为了探究沙棘多糖(Hippophae rhamnoides L.polysaccharide,HRP)对绵羊精液冷冻保存效果的影响,试验采集3 只湖羊种公羊精液,将其混合后随机分为 5 组,其中以未添加 HRP 的 Optidyl 稀释液进行等温稀释的作为对照组和鲜精组,其余 3 组分别用添加了终质量浓度为 5,10,15 g/L HRP 的 Optidyl 稀释液等温稀释精液,测定冻融精子活力、运动学参数[移动角度(MAD)、直线速度(VSL)、直线性(LIN)、前向性(STR)]、顶体完整率和质膜完整率,筛选出最适浓度后检测氧化应激指标[总超氧化物歧化酶(T-SOD)、谷胱甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC)、过氧化氢酶(CAT)、丙二醛(MDA)]、ATP 含量、线粒体活性及 Ca2+浓度.结果表明:10 g/L HRP 组的精子活率显著高于 15 g/L HRP 组(P<0.05);精子活力、A+B 精子数、MAD、LIN、STR 均显著高于对照组、5 g/L HRP 组和15 g/L HRP 组(P<0.05);VSL 显著高于对照组和5 g/L HRP 组(P<0.05);精子顶体完整率显著高于对照组和15 g/L HRP 组(P<0.05);质膜完整率显著高于其他组(P<0.05).10 g/L HRP 组 T-SOD 活性、CAT 活性、T-AOC、ATP 含量显著高于对照组(P<0.05),MDA 含量显著低于对照组(P<0.05);T-SOD、GSH-Px 活性、MDA、ATP 含量与鲜精组差异不显著(P>0.05),CAT 活性、T-AOC 显著低于鲜精组(P<0.05);线粒体活性、Ca2+浓度显著高于对照组(P<0.05),低于鲜精组,但差异不显著(P>0.05).说明在绵羊精液冷冻稀释液中添加 10 g/L HRP 可提高冻融精子活力、生物膜功能和抗氧化能力.

In order to explore the effects of Hippophae rhamnoides L.polysaccharide(HRP)on the cryopreservation of sheep semen,the semen of three Hu sheep rams was collected and randomly divided into five groups after mixing.The control group and fresh semen group were isothermally diluted without HRP Optidyl diluent,and the other three groups were isothermally diluted with Optidyl diluent with final mass concentrations of 5,10,and 15 g/L HRP,respectively.The motility,kinematics parameters,acrosome integrity rate and plasma membrane integrity rate of frozen-thawed sperm were measured.After screening the optimal concentration(moving angle[MAD],linear relocity[VSL],linearity[LIN],forwardness[STR]),oxidative stress indexes(total superoxide dismutase[T-SOD],glutathione peroxidase[GSH-Px],total antioxidant capacity[T-AOC],catalase[CAT],malondialdehyde[MDA]),ATP content,mitochondrial activity and Ca2+concentration were detected.The results showed that the sperm motility of 10 g/L HRP group was significantly higher than that of 15 g/L HRP group(P<0.05).Sperm motility,A+B sperm count,MAD,LIN and STR were significantly higher than those in the control group,5 g/L HRP group and 15 g/L HRP group(P<0.05).VSL was significantly higher than that in the control group and 5 g/L HRP group(P<0.05).The acrosome integrity rate of sperm was significantly higher than that of control group and 15 g/L HRP group(P<0.05).The plasma membrane integrity rate was significantly higher than that of other groups(P<0.05).T-SOD,CAT activity,T-AOC and ATP content in 10 g/L HRP group were significantly higher than those in the control group(P<0.05),and MDA content was significantly lower than that in the control group(P<0.05).There was no significant difference in T-SOD,GSH-Px activity and MDA-ATP content between 10 g/L HPR group and fresh semen group(P>0.05),while CAT activity and T-AOC were significantly lower than those in fresh semen group(P<0.05).Mitochondrial activity and Ca2+concentration were significantly higher than the control group(P<0.05),lower than the fresh sperm group,but the difference was not significant(P>0.05).The results indicated that adding 10 g/L HRP to sheep semen frozen diluent could improve the motility,biofilm function and antioxidant capacity of frozen-thawed sperm.
