绞股蓝皂苷对沂蒙黑猪精液冷冻保存效果的影响OA
Effects of Gynostemma pentaphyllum saponins on the preservation of frozen semen of Yimeng black pigs
为了探究不同浓度的绞股蓝皂苷(Gynostemma pentaphyllum saponins,GPS)对沂蒙黑猪冷冻精液保存效果的影响,试验将 12 头沂蒙黑猪的精液混合后随机分为对照组、低剂量组(GL 组)、中剂量组(GM 组)、高剂量组(GH 组),其中对照组中不添加 GPS,其余 3 组分别添加终质量浓度为 0.2,0.5,1 g/L GPS,测定精液解冻后精子活力及精子直线速度(VSL)、精子平均路径速度(VAP)、精子直线性(LIN)、精子前向性(STR)、精子摆动性(WOB)、质膜损伤率、DNA 损伤率、总抗氧化能力(T-AOC)、丙二醛(MDA)浓度、超氧化物歧化酶(SOD)活性、线粒体膜电位、活性氧(ROS)水平,检测 HK2、SOD1 和 BCL-2 基因 mRNA 的相对表达量.结果表明:与对照组相比,GL 组和 CM 组精子活力、VSL、VAP、LIN、STR、WOB、线粒体膜电位水平、SOD 活性、T-AOC 及HK2、BCL-2 基因mRNA 的相对表达量极显著升高(P<0.01),SOD1 基因mRNA 的相对表达量显著或极显著升高(P<0.05 或P<0.01),ROS 水平、质膜损伤率、DNA损伤率、MDA 浓度显著或极显著下降(P<0.05 或 P<0.01);而 GH 组精子活力、VSL、VAP、LIN、STR、线粒体膜电位水平、SOD 活性、T-AOC 及 SOD1、HK2 基因 mRNA 的相对表达量极显著降低(P<0.01),ROS 水平、质膜损伤率、DNA 损伤率、MDA 浓度极显著升高(P<0.01).说明在一定浓度范围内,GPS 对沂蒙黑猪精液质量提升作用呈现明显的剂量依赖效应.
In order to explore the effects of different concentrations of Gynostemma pentaphyllum saponins(GPS)on the preservation of frozen semen of Yimeng black pigs,the semen of 12 Yimeng black pigs were mixed and randomly divided into the control group,low dose group(GL group),middle dose group(GM group)and high dose group(GH group).The control group was not added with GPS,and the other three groups were added with GPS at a final mass concentration of 0.2,0.5,and 1 g/L,respectively.The sperm motility,sperm linear velocity(VSL),sperm average path velocity(VAP),sperm linear(LIN),sperm forward(STR),sperm wobble(WOB),plasma membrane damage rate,DNA damage rate,total antioxidant capacity(T-AOC),malondialdehyde(MDA)content,superoxide dismutase(SOD)activity,mitochondrial membrane potential,and reactive oxygen species(ROS)level were measured after semen thawing.The relative expression levels of HK2,SOD1 and BCL-2 mRNA were detected.The results showed that compared with the control group,the sperm motility,VSL,VAP,LIN,STR,WOB,mitochondrial membrane potential,SOD activity,T-AOC,and the relative expression of HK2 and BCL-2 mRNA in GL group and CM group were significantly increased(P<0.01),and the relative expression of SOD1 mRNA was significantly increased(P<0.05 or P<0.01).The levels of ROS,plasma membrane damage rate,DNA damage rate and MDA concentration were significantly or extremely significantly decreased(P<0.05 or P<0.01).The sperm motility,VSL,VAP,LIN,STR,mitochondrial membrane potential level,SOD activity,T-AOC,SOD1,HK2 gene mRNA relative expression in GH group were significantly decreased(P<0.01);ROS level,plasma membrane damage rate,DNA damage rate and MDA concentration were significantly increased(P<0.01).It indicated that GPS had a significant dose-dependent effect on the improvement of semen quality of Yimeng black pigs in a certain concentration range.
