垂盆草-白英提取工艺优选及活性成分含量测定研究OA
Optimization of extraction process and determination of active components in Sedum sarmentosum-Solanum lyratum Herb Pair
垂盆草-白英水煎剂源于古方"千家妙方"用于治疗肺癌,为提高该药对有效成分的提取效率,采用回流提取法,对水提工艺与醇提工艺进行比较与优化.以出膏量为初步考察指标,在单因素试验的基础上,建立高效液相色谱法(HPLC)对提取物中黄酮类成分含芦丁的量进行准确测定与对比,以考察不同工艺对抗肿瘤有效成分的提取效果.结果表明,水提法的最佳条件为料液比1∶10、90℃提取40 min;醇提法的最佳条件为乙醇体积分数80%、料液比1∶10、80℃提取90 min.两种方法下,醇提物和水提物中含芦丁的平均量分别为1.787、0.482 μg/g,各项方法学考察 RSD 值均小于5%.80%乙醇回流工艺的提取结果显著优于水提法,且醇提物中抗肿瘤活性成分的含量显著高于水提物,为垂盆草-白英药对的深度开发与利用提供实验基础.
The water decoction of the Sedum sarmentosum-Solanum lyratum herb pair originates from the ancient prescription Qianjia Miaofang and is used to treat lung cancer.To improve the extraction efficiency of its active components,this study compared and optimized the water and ethanol extraction processes using the reflux extraction method.With the extraction yield as the preliminary evaluation index,based on single-factor experiments,a high performance liquid chromatography(HPLC)method was established to accurately determine and compare the content of the flavonoid rutin in the extracts,in order to investigate the extraction effect of different processes on anti-tumor active components.The results showed that the optimal conditions for water extraction were a solid-liquid ratio of 1∶10 and extracting at 90℃for 40 min;the optimal conditions for ethanol extraction were 80%ethanol,a solid-liquid ratio of 1∶10,and extracting at 80℃for 90 min.Under the two methods,the average contents of rutin in the ethanol and water extracts were 1.787 μg/g and 0.482 μg/g,respectively,and the RSD values of all methodological validations were less than 5%.The extraction results of the 80%ethanol reflux process were significantly better than those of the water extraction method,and the content of anti-tumor active components in the ethanol extract was significantly higher than that in the water extract.This study provided an experimental basis for the in-depth development and utilization of the Sedum sarmentosum-Solanum lyratum herb pair.
