Bidirectional causal association between gut microbiota and Alzheimer's disease based on Mendelian randomization

目的:探讨肠道菌群(GM)与阿尔茨海默症(AD)之间潜在的因果关系.方法:收集 473 种 GM 的全基因组关联研究(GWAS)汇总数据,以及芬兰数据库 R12 中 AD 的 GWAS 汇总数据.以473 种 GM 的单核苷酸多态性(SNP)作为暴露,AD 为结局进行正向孟德尔随机化(MR)分析;以 AD 相关 SNP 作为暴露,将正向 MR 分析筛选出的菌群作为结局进行反向 MR 分析.结果:正向 MR 的逆方差加权(IVW)法结果显示,蓝藻门[OR(95%CI)为1.241(1.006~1.531)]、厚壁菌门-CAG-110[1.115(1.003~1.240)]、弗朗西斯菌目[1.594(1.121~2.266)]、毛螺菌科-CAG-488、优杆菌属、克雷伯菌属、固氮根瘤菌属、假链状双歧杆菌种、GCA-900066135 种 sp900066135 与 AD呈正关联,副衣原体目[OR(95%CI)为 0.586(0.356~0.967)]、孢子囊菌目[0.652(0.428~0.993)]、毛螺菌科-CAG-449、脱硫弧菌科、螺孢菌属、马赛菌属 sp002160815 种与 AD 呈负关联.反向 MR 分析结果显示,上述 15 种GM 与 AD 无反向因果关系.结论:部分 GM 与 AD 存在因果关联.

Aim:To investigate the potential causal relationship between gut microbiota(GM)and Alzheimer's disease(AD).Methods:Genome-wide association study(GWAS)data of 473 GM variants,as well as GWAS of AD from the Finn-ish R12 database were collected.Using single nucleotide polymorphisms(SNPs)of these 473 GM variants as exposures and AD as the outcome variable,forward Mendelian randomization(MR)analysis was conducted;using AD-associated SNPs as exposures and the microbial communities identified by forward MR analysis as the outcome variable,reverse MR analysis was performed.Results:The results of the inverse variance weighting(IVW)method for forward MR indicated that cya-nobacteria,and Francisellales Firmicutes-CAG-110[OR(95%CI)was 1.241(1.006-1.531)],[1.115(1.003-1.240)],[1.594(1.121-2.266)],Lachnospiraceae-CAD-488,Eubacterium,Klebsiella,Azorhizobium,Pseudocatenibacte-rium sp,GCA-900066135,sp900066135 were positively correlated with AD.There was a negative correlation between AD and sp002160815 species in the order arechlamydiales[OR(95%CI)was 0.586(0.356-0.967)],sporangiales[0.652(0.428-0.993)],family Trichomycteridae-CAG-449,family Desulfovibrio,genus Spirosporium,and genus Maasai.Re-verse MR analysis revealed no reverse causal relationship between the aforementioned 15 GM species and AD.Conclusion:Partial GM may be causal associated with AD.
