目的:分析肠激安方(CJA)对脂多糖致炎人结肠上皮细胞 NCM460 细胞活力、NF-κB/肌球蛋白轻链激酶(MLCK)通路和紧密连接蛋白表达的影响.方法:将 NCM460 细胞分为 4 组.正常对照组用含体积分数 10%胎牛血清的培养基培养,模型组用含体积分数10%胎牛血清的培养基和100 ng/mL 脂多糖培养,空白血清组用含体积分数10%空白血清的培养基和100 ng/mL 脂多糖培养,含药血清组用含体积分数10%CJA 含药血清的培养基和100 ng/mL 脂多糖培养.24 h 后应用 CCK-8 法检测细胞活力,RT-qPCR 法检测 NF-κB/MLCK 通路相关 mRNA 的表达,Western blot 法检测 NF-κB/MLCK 通路相关蛋白及紧密连接蛋白的表达.结果:与正常对照组比较,模型组和空白血清组细胞活力、肿瘤坏死因子受体相关因子(TRAF3)mRNA 及蛋白表达水平、紧密连接蛋白表达水平降低;TNF-α、MLCK mRNA 和蛋白表达水平升高(P<0.05),p65 mRNA 及 p65、肌球蛋白轻链(MLC)蛋白磷酸化水平升高(P<0.05).与模型组和空白血清组比较,CJA 含药血清组细胞活力恢复,TRAF3 mRNA 与蛋白表达水平、紧密连接蛋白表达水平升高(P<0.05);TNF-α、MLCKmRNA 与蛋白,p65 mRNA 及 p65、MLC 蛋白磷酸化水平降低(P<0.05).结论:CJA 可抑制 NF-κB/MLCK 通路激活,改善脂多糖致炎 NCM460 细胞活力,可能参与改善结肠上皮细胞膜的通透性.

Aim:To investigate the effects of Changji'an Formula(CJA)on the viability of LPS-induced NCM460 cells and molecules involved in the NF-κB/MLCK pathway and the expression of tight junction proteins.Methods:NCM460 cells were divided into 4 groups.The normal control group was cultured in medium containing 10%(V/V)fetal bovine serum(FBS).The model group was cultured in medium containing 10%(V/V)FBS supplemented with 100 ng/mL lipopolysac-charide(LPS).The blank serum group was cultured in medium containing 10%(V/V)blank serum and 100 ng/mL LPS.The medicated serum group(CJA group)was cultured in medium containing 10%(V/V)CJA-serum and 100 ng/mL LPS.After 24 hours of incubation,cell viability was assessed using CCK-8 assay.The mRNA expression of NF-κB/MLCK path-way-related genes were determined by RT-qPCR,the expression of NF-κB/MLCK pathway-related proteins and tight junc-tion proteins were detected by Western blot.Results:Compared with the normal control group,the model group and blank serum group showed significantly reduced cell viability,decreased mRNA and protein expression levels of TRAF3,and re-duced expression of tight junction proteins;while the mRNA and protein expression levels of TNF-α and MLCK,as well as the expression levels of p-p65/p65 and p-MLC/MLC were significantly increased(P<0.05).Compared with the model group and blank serum group,CJA group showed significantly restored cell viability,increased TRAF3 mRNA and protein expression,reduced mRNA and protein expression levels of TNF-α,MLCK,p65 mRNA expression as well as the p65 mRNA expression level,as well as the expression levels of p-p65/p65 and p-MLC/MLC,and restored the expression levels of tight junction proteins(P<0.05).Conclusion:CJA can inhibit activation of NF-κB/MLCK pathway,improve the viability of NCM460 cells,and may contribute to the amelioration of membrane permeability.
