淫羊藿苷对骨质疏松大鼠种植体骨结合及骨相关基因血管内皮生长因子与组织形态学的影响OA
Effects of Icariin on Implant Osseointegration Bone-related Genes Vascular Endothelial Growth Factor and Histomorphology in Osteoporosis Rats
目的:研究骨质疏松大鼠经淫羊藿苷干预后的作用机制,以及对种植体骨结合及骨相关基因、血管内皮生长因子与组织形态学的影响.方法:选取 26 只 SD 清洁级雌性大鼠,随机分为假手术组、模型组、淫羊藿苷组,每组各8 只,其中包含2 只建模失败大鼠.对比各组植入物周围骨参数、种植体骨结合情况、骨组织形态学变化、骨代谢指标水平、氧化应激指标水平、成骨细胞凋亡率、骨相关基因、血管内皮生长因子表达.结果:与假手术组相比,模型组 GSH-Px、松质骨区骨量、CAT、Tb.Th、骨保护素、骨结合率、BMD、25OHD、骨钙素、Tb.N、OPG、骨桥蛋白、BV/TV、血管内皮生长因子、SOD 水平下降,ALP、凋亡率、Tb.Sp、MDA 水平升高;与模型组相比,淫羊藿苷组 GSH-Px、骨结合率、CAT、Tb.N、松质骨区骨量、BV/TV、25OHD、骨保护素、Tb.Th、OPG、骨钙素、BMD、骨桥蛋白、SOD、血管内皮生长因子水平上升,凋亡率、MDA、Tb.Sp、ALP 水平降低,差异具有统计学意义(P<0.05).结论:淫羊藿苷可改善骨质疏松大鼠血管内皮生长因子、种植体骨结合、骨相关基因表达情况,降低成骨细胞凋亡率,调节氧化应激及骨代谢水平.
Objective:To investigate the action mechanism of icariin intervention in osteoporotic rats,as well as its effects on implant osseointegration,bone-related genes,vascular endothelial growth factor(VEGF),and histomorphology.Methods:Twenty-six SD clean-grade female rats were selected and ran-domly divided into the sham operation group,the model group,and the icariin group,with 8 rats in each group,excluding 2 rats with failed modeling.The bone parameters around the implants,implant osseointegra-tion,bone tissue morphological changes,bone metabolism index levels,oxidative stress index levels,osteo-blast apoptosis rate,bone-related gene expression,and VEGF expression were compared among the three groups.Results:Compared with the sham operation group,the model group showed significant decreases in the levels of GSH-Px,bone mass in the cancellous bone area,CAT,Tb.Th,osteoprotegerin,osseointegra-tion rate,BMD,25OHD,osteocalcin,Tb.N,OPG,osteopontin,BV/TV,VEGF,and SOD;meanwhile,the levels of ALP,apoptosis rate,Tb.Sp,and MDA were significantly increased(all P<0.05).Compared with the model group,the icariin group exhibited significant increases in the levels of GSH-Px,osseointegra-tion rate,CAT,Tb.N,bone mass in the cancellous bone area,BV/TV,25OHD,osteoprotegerin,Tb.Th,OPG,osteocalcin,BMD,osteopontin,SOD,and VEGF,while the apoptosis rate,MDA,Tb.Sp,and ALP levels were significantly decreased(all P<0.05).Conclusions:Icariin can improve VEGF expression,im-plant osseointegration,and bone-related gene expression in osteoporotic rats,reduce the osteoblast apoptosis rate,and regulate oxidative stress and bone metabolism levels.
