Optimization of extraction process of Xiao'er Qushi Zhiyang Granules based on AHP-EWM combined with response surface methodology

为提高小儿祛湿止痒颗粒有效成分的提取率并增加制剂质量的稳定性,采用单因素实验结合Box-Behnken响应面法对其水提取工艺进行优化.以加水倍数、提取时间、浸泡时间为考察因素,以迷迭香酸、芍药苷、黄柏酮的含量及干膏率为评价指标,采用层次分析法-熵权法(analytic hierarchy process-entropy weight method,AHP-EWM)组合赋权计算综合评分,然后通过响应面法分析因素交互作用并优选工艺参数,同时开展方法学考察与工艺验证.结果表明:最优水提工艺为加8倍量水,浸泡30 min,分2次提取,总提取时长为3 h;该工艺下的综合评分均值为95.80,与预测值的相对误差小于1.0%.优化后的水提取工艺稳定可行、重现性好,可充分提取方中迷迭香酸、芍药苷、黄柏酮等核心药效成分,可为小儿祛湿止痒颗粒的工业化生产与质量控制提供参考.

To improve the extraction yield of effective components from Xiao'er Qushi Zhiyang Granules and enhance the stability of preparation quality,this study optimizes the water extraction process of Xiao'er Qushi Zhiyang Granules by combining single-factor experiments with the Box-Behnken response surface methodology.The amount of added water,extraction time and soaking time were selected as investigation factors,while the contents of rosmarinic acid,paeoniflorin and obacunone and the yield of dry extract were adopted as evaluation indicators.A combined weighting method of Analytic Hierarchy Process-Entropy Weight Method(AHP-EWM)was applied to calculate the comprehensive score.Response surface methodology was employed to analyze the interaction between factors,optimize process parameters,and carry out methodological research and process validation.The results show that the optimal water extraction process is as follows:adding eight times the amount of water,soaking for 30 minutes,extracting two times,with a total extraction time of three hours.Under this process,the comprehensive score is 95.80 with RSD=0.96%,and the relative error compared with the predicted value is less than 1.0%.In conclusion,this process is stable,feasible and has good reproducibility,which can sufficiently extract paeoniflorin,rosmarinic acid,obacunone and other core pharmacodynamically active components in the formula,and can provide a reference for the industrial production and quality control of Xiao'er Qushi Zhiyang Granules.
