外源咖啡酸处理对中国沙棘种子萌发的影响OA
Effects of Exogenous Caffeic Acid on Seed Germination of Hippophae Rhamnoides subsp.Sinensis Rousi
为探究外源咖啡酸对中国沙棘种子萌发的适宜浓度及作用机制,以兴隆山中国沙棘种子为试验材料,研究不同浓度梯度(0、25、50、100、200、400mg/L)咖啡酸处理对中国沙棘种子萌发的影响.结果显示,外源咖啡酸对中国沙棘种子萌发存在"低促高抑"的浓度效应.低浓度(25、50mg/L)在萌发关键期(第5天至第7天)有促进效果,25 mg/L咖啡酸处理在第5天的发芽势最高,萌发整齐度和胚根生长突出;50mg/L处理则能同步提高发芽率、发芽势与鲜重,缩短平均萌发时间,两个浓度均能激活抗氧化系统,增强过氧化物酶活性并维持较低丙二醛含量.高浓度(200、400 mg/L)咖啡酸抑制种子萌发,关键萌发指标全面下降,过氧化物酶活性受抑、丙二醛含量急剧上升.除发芽第6天50 mg/L处理与ck有显著差异外,其余处理对种子干重无显著影响.综上所述,25、50mg/L浓度的外源咖啡酸通过增强抗氧化活性、缓解氧化损伤促进种子萌发,为沙棘育苗中外源化学物质调控提供依据.
To investigate the optimal concentration and underlying mechanism of exogenous caffeic acid on seed germination of Hippophae rhamnoides subsp.Sinensis Rousi,seeds collected from Xinglong Mountain were treated with different concen-trations(0,25,50,100,200,400 mg/L)of caffeic acid.The results showed that exogenous caffeic acid exhibited low-concentration promotion and high-concentration inhibition effect on seed germination.Low concentrations(25 mg/L and 50 mg/L)promoted germination during the critical period(days 5 to 7).The 25 mg/L treatment yielded the highest germination potential on day 5,with notable improvements in germination uniformity and radicle growth.The 50 mg/L treatment simulta-neously increased germination rate,germination potential,and fresh weight,while reducing the mean germination time.Both concentrations activated the antioxidant system,enhanced peroxidase activity,and maintained low malondialdehyde content.High concentrations(200 mg/L and 400 mg/L)inhibited seed germination,leading to a comprehensive decline in key germi-nation indices,suppressed peroxidase activity,and a sharp increase in malondialdehyde content.Except for the 50 mg/L treatment on day 6,which showed a significant difference from the control,no significant effects on seed dry weight were ob-served for other treatments.In conclusion,exogenous caffeic acid at concentrations of 25 mg/L and 50 mg/L promoted seed germination by enhancing antioxidant activity and alleviating oxidative damage,providing a basis for the application of exog-enous chemicals in seabuckthorn seedling cultivation.
