Identification of Genetic Loci for Leaf Shape Traits Based on Backcross Population of Wild Soybean

为挖掘和丰富调控大豆叶形性状遗传位点,本研究以栽培品种冀豆 12 为轮回亲本,野生大豆ZYD2738 为供体亲本,构建了3 个共计包含168 个家系的野生大豆回交群体,进行大豆亲本间叶形表型差异分析、群体间叶形表型变异和遗传分析及大豆叶形QTL定位,挖掘调控大豆叶形的遗传位点,探究大豆叶形的遗传机制.表型变异和遗传分析结果显示:与 ZYD2738 相比,冀豆 12 的左叶长、中叶长、右叶长、左叶宽、中叶宽和右叶宽分别显著增加399.59%、374.94%、431.53%、576.39%、531.90%和 567.42%(P<0.001);而冀豆 12 左叶夹角的角度显著低于ZYD2738,为其86.77%(P<0.01);除叶夹角外,其他叶形性状的遗传力在0.55 以上.双因素方差分析结果显示,环境、基因型和环境×基因型对大豆株型性状影响的F值范围分别为0.13~5 556.15、5.97~19.11 和4.90~21.33,表明两种环境下,环境、基因型、环境×基因型均显著影响大豆的叶形性状(P<0.001).基于重测序技术绘制了3 个野生大豆回交群体物理指纹图谱,14006、14008 和14022 群体分别含有 997,996 和 990 个标记.QTL定位结果显示,共定位到47 个叶形相关QTLs,LOD值为2.50~5.45,可解释表型变异11.53%~32.20%.新鉴定到3 个调控大豆叶形的共定位遗传位点,加性效应显示Locus4_2 和Locus17 的功能基因来源于冀豆12,Locus9_1 来源于ZYD2738.研究结果为充分挖掘和利用野生大豆资源和进一步探究大豆叶形遗传调控机制提供了遗传信息和材料.

To identify and enrich genetic loci regulating soybean leaf shape traits,three wild soybean backcross populations consisting of 168 lines in total were developed using the cultivated soybean cultivar Jidou 12 as the recurrent parent and wild soybean ZYD2738 as the donor parent in this study.Phenotypic differences in leaf shape between the two parents,phenotypic variation and genetic analysis of leaf shape across populations,and QTL mapping for soybean leaf shape were performed to mine genetic loci and dissect the genetic mechanisms underlying soybean leaf shape.Phenotypic variation and genetic analyses revealed that compared with ZYD2738,Jidou 12 exhibited significantly increased left leaf length,middle leaf length,right leaf length,left leaf width,middle leaf width and right leaf width by 399.59%,374.94%,431.53%,576.39%,531.90%and 567.42%,respectively(P<0.001).The left leaf angle of Jidou 12 was significantly smaller,accounting for 86.77%of that of ZYD2738(P<0.01).Except for leaf angle,the heritability of all other leaf shape traits was higher than 0.55.Two-way ANOVA showed that the F-values of environment,genotype and genotype-by-environment interaction(G×E)for soybean leaf shape traits ranged from 0.13 to 5 556.15,5.97 to 19.11,and 4.90 to 21.33,respectively.These results indicated that environment,genotype and G×E interaction all exerted significant effects on soybean leaf shape traits across the two environments(P<0.001).Physical fingerprint maps of the three wild soybean backcross populations were constructed based on resequencing technology,with 997,996 and 990 molecular markers detected in populations 14006,14008 and 14022,respectively.A total of 47 QTLs associated with leaf shape were identified,with LOD scores ranging from 2.50 to 5.45 and the phenotypic variation explained ranging from 11.53%to 32.20%.Among these,three novel co-localized genetic loci governing soybean leaf shape were newly identified.Analysis of additive effects demonstrated that the functional genes of Locus4_2 and Locus17 were derived from Jidou 12,while Locus9_1 originated from ZYD2738.In conclusion,this study provides valuable genetic resources and foundational data for the exploration and utilization of wild soybean germplasms,as well as further investigations into the genetic regulatory mechanisms of soybean leaf shape.
