为全面探究大豆NF-YA基因家族在大豆生长发育中的作用及其对油分含量的影响,本研究利用生物信息学和分子生物学方法,鉴定了21 个大豆NF-YA家族基因,分别命名为GmNFYA1~GmNFYA21.分子生物学特征预测表明,NF-YA蛋白质长度为205~346 aa,分子量为22.87~37.91 kD,等电点范围为 5.73~9.98,均为带负电荷的亲水性蛋白,亚细胞预测定位显示大多数NF-YA(GmNFYA12、GmNFYA19 和GmNFYA21 除外)定位于细胞核.染色体分布和共线性分析结果表明,NF-YA基因家族成员分布在15 条染色体上,该家族有28 对基因具有共线性关系,呈现明显的功能分化趋势.基因表达模式分析表明GmNFYA2、GmNFYA11、GmNFYA14、GmNFYA17 在绥农 14 和ZYD00006的EM1 时期的籽粒存在表达差异.基于547 份大豆种质的单倍型分析结果表明,GmNFYA2 和GmNFYA14 存在油分含量优异单倍型.本研究为全面解析NF-YA家族基因调控大豆油分合成与代谢的生物学功能提供了重要线索.

To comprehensively explore the functions of the soybean NF-YA gene family in soybean growth and development as well as its effects on seed oil content,in this study,21 soybean NF-YA family genes were identified using bioinformatics and molecular biology methods and named GmNFYA1-GmNFYA21.Molecular biological characteristic prediction indicated that NF-YA proteins ranged in length from 205 to 346 amino acids,with molecular weight between 22.87 and 37.91 kD and isoelectric point ranging from 5.73 to 9.98.All were negatively charged hydrophilic proteins,and most NF-YAs(except GmNFYA12,GmNFYA19,and GmNFYA21)were located in the nucleus.Chromosome distribution and collinearity analysis results showed that NF-YA gene family members were distributed on 15 chromosomes,and 28 pairs of genes in this family had collinearity relationships,showing a clear trend of functional differentiation.Gene expression pattern analysis indicated that GmNFYA2,GmNFYA11,GmNFYA14,and GmNFYA17 had expression differences in the EM1 stage of seeds in Suinong 14 and ZYD00006.Haplotype analysis based on 547 soybean germplasms showed that GmNFYA2 and GmNFYA14 had superior haplotypes for oil content.This study provides important clues for comprehensively understanding the biological functions of NF-YA family genes in regulating soybean oil synthesis and metabolism.
