三黄泻心汤胃内滞留片的制备工艺及质量评价的初步研究OA
Research on Preparation Process and Preliminary Quality Evaluation of Sanhuang Xiexin Gastric Retention Tablets
目的:本研究旨在制备一种三黄胃内滞留片,将三黄泻心汤中的有效成分通过现代制剂技术制备成固体剂型,保留其药效,同时实现缓释和胃内滞留的效果,从而增强治疗胃溃疡的作用.方法:本研究以粉末直接压片法进行片剂的制备,并采用单因素试验来考察骨架材料、助漂剂和起泡剂的种类及用量,以漂浮性能和累积释放度作为评价指标.采用正交试验进一步优化处方,对制备的三黄胃内滞留片进行质量初步评价和释药机制的研究.结果:三黄胃内滞留片的最优处方为:HPMC K15M用量为30%,NaHCO3用量为5%,十八醇用量为15%,微晶纤维素用量为5%,聚乙二醇6000用量为5%,硬脂酸镁用量为1%,三黄BBT混相体用量为39%,片重为0.35 g.三黄胃内滞留片在一分钟内均可起漂,平均漂浮时间长达12 h,具有明显的缓释特征,12 h的累积释放度可达80%以上,体外释药过程符合Ritger-Peppas模型,释药机理为扩散与溶蚀协同.结论:制备得到的三黄胃内滞留片,浮动滞后时间较短,持续漂浮时间较长,缓释作用明显.
Objective:This study aims to develop a Sanhuang gastric retention tablet.By employing modern formulation technology,the active components of Sanhuang Xiexin Decoction are prepared into a solid dosage form.This process preserves the efficacy while achieving sustained release and gastric retention,thereby enhancing its therapeutic effect on gastric ulcers.Methods:In this study,these tablets were prepared by direct powder compression.A single-factor test was employed to investigate the type and amount of matrix materials,floating aid agents,and foaming agents,using floating performance and cumulative drug release as evaluation indica-tors.An orthogonal experimental design was further applied to optimize the formulation.Preliminary quality evaluation and a study on the drug release mechanism of the prepared Sanhuang gastric-retentive tablets were conducted.Results:The optimal formulation was determined as follows:30%HPMC K15M,5%NaHCO3,15%stearyl alcohol,5%microcrystalline cellulose,5%polyethylene gly-col 6000,1%magnesium stearate,and 39%Sanhuang drug.The tablet's weight was 0.35 g.The prepared Sanhuang gastric-retentive tablets exhibited buoyancy within one minute,with an average sustained floating duration of 12 h.Moreover,they demonstrated signifi-cant sustained-release characteristics,achieving a cumulative drug release of over 80%at 12 h.The in vitro drug release profile fitted the Ritger-Peppas model,indicating a release mechanism combining diffusion and erosion.Conclusion:The developed Sanhuang gas-tric retentive tablets exhibit a short floating lag time,prolonged sustained floating duration,and pronounced sustained-release proper-ties.
