以22个肺形侧耳菌株为试材,采用最小显著差法和极差法确定测量性状的分级范围,通过变异系数、遗传多样性指数及聚类分析、相关性分析和主成分分析等方法评价遗传多样性,以期为肺形侧耳新品种选育提供有效的种质资源.结果表明:原基形成时间、菌盖长度等8个农艺性状符合正态分布,采用最小显著差法分成3级;发菌时间和菌盖厚度不符合正态分布,采用极差法分成3级;上述10个农艺性状在供试菌株间的变异系数范围在5.34%~29.63%.13个农艺性状的遗传多样性指数在0.49~1.39,其中菌盖颜色、原基形成时间、菌柄长度、菌盖菌柄长度比和菌柄形状的遗传多样性指数较大,表明这些性状的表型多样性较为丰富;原基形成时间与子实体成熟时间、菌盖长度与菌盖宽度、菌柄直径和菌盖厚度之间存在高度相关性.聚类分析结果表明,所有供试菌株能够划分成5个类群,栽培种质的农艺性状同质化现象严重,野生种质农艺性状多样性丰富.主成分分析结果表明,前3个主成分累计贡献率为71.750%,分别代表了菌盖特征、菌柄特征和发育周期.基于主成分分析结果对供试菌株进行综合评价,得分范围为-1.00~1.31,综合得分较高的5个肺形侧耳菌株可作为新品种选育的优异资源予以利用.中国肺形侧耳种质资源的表型多样性丰富,具有良好的驯化利用潜力.

Taking total of 22 Pleurotus pulmonarius strains as the test materials,the range of grades for the measured traits was determined by the least significant difference method and the range method.The genetic diversity was assessed by coefficient of variation,genetic diversity index,and clustering,correlation and principal component analysis,in order to provide effective germplasm resources for the breeding of new varieties.The results showed that 8 agronomic traits such as primordium formation time and pileus length conformed to normal distribution and were divided into three grades by the least significant difference method;while spawn running period and pileus thickness did not conform to normal distribution and were also divided into three grades by the range method.The coefficient of variation of the above 10 agronomic traits among the tested strains ranged from 5.34%to 29.63%.The genetic diversity index of 13 agronomic traits varied from 0.49 to 1.39.Among 13 agronomic traits,the genetic diversity indexes of pileus color,primordium formation time,stipe length,ratio of pileus length to stipe length and stipe shape were relatively large,indicated that the phenotypic diversity of these agronomic traits was relatively abundant.There was a high correlation between primordium formation time and fruiting body maturity time,as well as pileus length and pileus width,stipe diameter and pileus thickness.The clustering analysis showed that all tested strains could be divided into 5 groups.The agronomic traits of cultivated germplasm were highly homogenized,while the agronomic traits of wild germplasm were rich in diversity.Principal component analysis showed the first three principal components accounting for 71.750%of the cumulative contribution,representing pileus characteristics,stipe characteristics and reproduction period,respectively.The comprehensive evaluation of the tested strains was conducted based on principal component analysis.The comprehensive score of the tested strains range was from-1.00 to 1.31.The five strains with higher comprehensive scores can be utilized as available germplasm for breeding new varieties of P.pulmonarius.The phenotypic diversity of P.pulmonarius germplasm resource is abundant,which exhibits significant potential for domestication and utilization.
