镍胁迫对薏苡种子萌发及幼苗生理特性的影响OA

为探究镍胁迫对薏苡种子萌发及幼苗生理特性的影响,以浦薏6号为供试材料,设8个镍浓度处理,分别为0(CK)、2.5、5、10、15、20、25、30 mg/L,测定每个处理的种子发芽率、发芽势、发芽指数,幼苗的根长、芽长及丙二醛含量、过氧化氢酶活性等指标.结果表明,镍离子浓度为5 mg/L时,种子的发芽率、发芽势分别为94.67%、94.00%,略高于CK;当镍离子浓度≥10 mg/L时,薏苡种子的发芽势和发芽率随浓度的增加,整体呈逐渐下降趋势.镍胁迫下,薏苡的根长、芽长、活力指数、芽鲜重、芽干重、根鲜重及根干重均低于CK.镍离子浓度在5~20 mg/L时,与CK相比,其叶绿素含量有所升高,电导率有所下降,过氧化氢酶活性有所提高,超氧化物歧化酶活性有所下降.以上结果表明,镍胁迫对薏苡的生长具有明显的抑制作用,但一定浓度范围内(5~20 mg/L)的胁迫可刺激植株调动自身防御系统,从而增强其适应能力.本文为薏苡耐镍品种选育提供参考.

To investigate the effects of nickel(Ni)stress on seed germination and seedling physiological characteristics of Coix lacryma-jobi,the cultivar Puyi No.6 was used as the test material.8 Ni concentration treatments were set,namely 0(CK),2.5,5,10,15,20,25,and 30 mg/L.For each treatment,seed germination rate,germination energy,germination index,seedling root length,shoot length,malondialdehyde content,and catalase activity were measured.The results showed that at the Ni concentration of 5 mg/L,the germination rate and germination energy reached 94.67%and 94.00%,respectively,which were slightly higher than those of the CK.When the Ni concentration reached≥10 mg/L,the germination energy and germination rate of Coix lacryma-jobi seeds showed an overall declining trend with increasing concentration.Compared with the CK,the root length,shoot length,vigor index,shoot fresh weight,shoot dry weight,root fresh weight,and root dry weight of Coix lacryma-jobi under Ni stress were all lower than those of the CK.At Ni concentrations ranging from 5 to 20 mg/L,compared with the CK,the chlorophyll content increased,the electrical conductivity decreased,the catalase activity increased,while the superoxide dismutase activity decreased.These results indicate that Ni stress has an obvious inhibitory effect on the growth of Coix lacryma-jobi,but stress within a certain concentration range(5-20 mg/L)can stimulate the plant to activate its own defense system,thereby enhancing its adaptability.This study provides a reference for the breeding of Ni-tolerant Coix lacryma-jobi varieties.
