Diras2条件性敲除对小鼠食管上皮细胞增殖的影响OA
Effects of Diras2 conditional knockout on proliferation of mouse esophageal epithelial cells
目的:探讨小鼠食管上皮细胞的增殖与Diras2蛋白的关系,为食管癌的早期诊断和治疗提供新的靶标.方法:取Diras2条件性敲除鼠和对照鼠的食管组织,利用免疫组织化学染色法检测食管组织中的Diras2蛋白表达以验证其表达水平.HE染色后镜下观察两组食管的组织形态.采用免疫组织化学染色法检测两组食管组织中Ki67、pRaf1、pMEK1/2、pErk1/2等蛋白表达.结果:Diras2条件性敲除小鼠食管中Diras2蛋白表达水平明显低于对照组小鼠(P<0.01).HE染色显示两组食管的组织形态学未发生明显变化.免疫组织化学检测显示,Diras2条件性敲除后小鼠食管上皮中的Ki67阳性细胞比例降低(P<0.01).同时,pRaf1、pMEK1/2、pErk1/2等蛋白表达亦明显降低(P<0.01).结论:Diras2条件性敲除可能通过Raf/MEK/Erk信号通路抑制小鼠食管上皮细胞的增殖.
OBJECTIVE:To provide a new target for early diagnosis and treatment of esophageal cancer,Diras2 conditional knockout mice and control mice were used to investigate control of esophageal epithelial cell proliferation by Diras2.METHODS:Esophagus tissues of Diras2 conditional knockout and control mice were collected.Immunohistochemical(IHC)staining method was used to detect expression of Diras2 in esophageal tissues.HE staining was used to reveal organization forms of esophagus tissues.IHC staining was performed to detect expressions of Ki67,pRaf1,pMEK1/2,pErk1/2 protein in each group.RESULTS:Expression levels of Diras2 protein in the conditional knockout mice were significantly lower than that in control mice(P<0.01).HE staining showed no significant changes in the histomorphology of the esophagus in both groups.Immunohistochemical staining showed that the proportion of Ki67-positive cells in esophageal epithelium of the conditional knockout mice was reduced(P<0.01).At the same time,expressions of pRaf1,pMEK1/2 and pErk1/2 proteins were also significantly reduced(P<0.01).CONCLUSION:Conditional knockout of Diras2 inhibited proliferation of esophageal epithelial cells in mice by regulating the Raf/MEK/Erk signaling pathway.
