食管癌中Z-DNA结合蛋白1与细胞焦亡相关分子干扰素调节因子3的相关性及其机制OA
Correlation analysis of ZBP1 and pyroptosis related molecule IRF3 in esophageal cancer
目的:探讨食管癌中Z-DNA结合蛋白1(ZBP1)与细胞焦亡相关分子干扰素调节因子3(IRF3)之间的相关性及其机制.方法:通过STRING数据库分析ZBP1与细胞焦亡相关分子的蛋白互作网络,利用GEPIA数据库分析食管癌中ZBP1与细胞焦亡相关分子的相关性,筛选相关指数最大的蛋白.采用免疫组织化学检测IRF3在食管癌组织中的表达.在食管癌Eca109细胞中构建ZBP1过表达细胞模型,并用Western blot法检测ZBP1与IRF3蛋白在ZBP1过表达细胞模型中的表达水平.结果:通过STRING及GEPIA数据库筛选,ZBP1在食管癌中的表达与细胞焦亡相关分子GSDMD、NLRP3、IRF3和TBK1(r<0.24,P<0.01)等的表达呈正相关,相互作用指数为IRF3>TBK1>TBK1>GSDMD,IRF3关联性最强,因此后续研究将以IRF3作为目标分子.免疫组织化学法检测显示IRF3表达水平在食管癌组织中的表达显著高于正常组织(P<0.05),但其表达水平与食管癌预后无明显关系(P>0.05).在ZBP1过表达食管癌细胞模型中,ZBP1表达明显增加,表明模型构建成功.Western blot检测结果表明ZBP1过表达细胞中IRF3蛋白的表达显著高于对照组(P<0.01).结论:IRF3在食管癌中的表达高于正常组织,其高表达可能与ZBP1的靶向调控有关.
OBJECTIVE:To investigate correlations and underlying mechanisms between Z-DNA binding protein 1(ZBP1)and interferon regulatory factor 3(IRF3),a molecule associated with pyroptosis,in esophageal carcinoma(EC).METHODS:The protein-protein interaction network of ZBP1 and pyroptosis-related molecules was analyzed using the STRING database.Correlations between ZBP1 and pyroptosis-related molecules in EC were assessed via the GEPIA database to screen proteins with the highest correlation coefficient.Immunohistochemistry was used to detect IRF3 expression in EC tissues.A ZBP1 overexpression model was constructed in Eca109 cells,and protein levels of ZBP1 and IRF3 were determined by Western blot.RESULTS:STRING and GEPIA analyses identified that IRF3 had the strongest correlation with ZBP1.Immunohistochemistry showed that IRF3 expression was significantly higher in EC tissues than in adjacent normal tissues(P<0.05),but was not associated with EC prognosis(P>0.05).The ZBP1 overexpression model was successfully verified by markedly increased ZBP1 expression.Western blot demonstrated that IRF3 protein expression was significantly elevated in ZBP1-overexpressing cells compared with the control group(P<0.01),suggesting that upregulated ZBP1 might promote IRF3 expression.CONCLUSION:IRF3 expression was higher in esophageal carcinoma than in normal tissues,and its upregulation may be regulated by ZBP1 in a targeted manner.
