基于生物信息学筛选类风湿性关节炎的生物标志物OA
Screening of rheumatoid arthritis biomarkers based on bioinformatics
为辅助诊断和治疗类风湿性关节炎(RA),本研究利用生物信息学方法,识别并验证了与 RA 相关的关键生物标志物.首先,从基因表达综合(GEO)数据库获取 RA 转录组数据集,分析筛选差异表达基因,并进行富集分析,探讨差异表达基因的生物学功能;然后,在蛋白-蛋白相互作用(PPI)网络中探讨 6 个关键基因(CCR7、CD2、CD27、CD8A、GZMB 和 IL7R),并使用独立的数据集和实时荧光定量聚合酶链式反应(RT-qPCR),验证基因的表达水平.研究表明,miR-9-5p 和 IL7R 是竞争性内源核糖核酸(RNA)网络的核心成分,并确定了 6 个关键基因,其中 IL7R 和 miR-9-5p 可能在 RA 的进展中发挥关键作用.本研究可为 RA 的诊疗提供新的方向.
To assist in the diagnosis and treatment of rheumatoid arthritis(RA),we employed bioinformatics approaches to identi-fy and validate key biomarkers associated with RA.Firstly,the transcriptomic dataset of RA was obtained from the Gene Expression Omnibus(GEO)database.Differentially expressed genes were analyzed and screened,followed by functional enrichment analysis to in-vestigate their biological roles.Subsequently,six hub genes(CCR7,CD2,CD27,CD8A,GZMB and IL7R)were identified in the protein-protein interaction(PPI)network,and the expression levels of these genes were verified using independent datasets and real-time quantitative polymerase chain reaction(RT-qPCR).The study demonstrated that miR-9-5p and IL7R were the core components of a competing endogenous ribonucleic acid(CeRNA)network,and six key genes were identified.Among them,IL7R and miR-9-5p might play a crucial role in the progression of RA.This research can provide a new direction for the diagnosis and treatment of RA.
