Differential Regulation of the Hippo Signaling Pathway in Heart Failure and Research Advances
心力衰竭是全球面临的重大健康问题,其发病率仍在逐年上升.Hippo信号通路作为调控细胞增殖、凋亡、机械转导及组织稳态的重要信号通路,在心力衰竭的发生发展过程中发挥着关键作用.本文系统梳理了Hippo信号通路核心分子的调控机制,重点探讨了该通路在不同心力衰竭亚型的阶段性变化以及在不同心脏细胞类型中的细胞特异性作用.同时,阐述了Hippo通路通过调节心肌细胞代谢与增殖、心脏成纤维细胞分化、内皮细胞功能及对巨噬细胞的差异性调控,从而参与心力衰竭的多种关键病理过程.此外,本文还对多种Hippo通路调节剂进行了梳理,关注其与传统心力衰竭治疗药物的区别及协同作用.本文总结了Hippo信号通路在心力衰竭过程中的具体分子机制,并为心力衰竭的治疗提供了新视角.
Heart failure is a major global health challenge,and its prevalence continues to rise.The Hippo pathway is a key regulator of cell proliferation,apoptosis,mechanotransduction,and tissue homeostasis,and it plays a critical role in the development and progression of heart failure.This review systematically summarizes the regulatory mechanisms of the core components of the Hippo pathway,with emphasis on its stage-specific alterations across different heart failure subtypes and its cell-specific roles in various car-diac cell populations.It further describes how the Hippo pathway is deeply involved in multiple key pathological processes of heart fail-ure through its effects on cardiomyocyte metabolism and proliferation,cardiac fibroblast differentiation,endothelial function,and the differential regulation of macrophage function.In addition,this review summarizes a range of Hippo pathway modulators,highlighting their differences from conventional heart failure therapies as well as their potential synergistic effects.Overall,this article outlines the specific molecular mechanisms of the Hippo pathway in heart failure and offers a new perspective for the treatment of this disease.
