河北地区番茄潜叶蛾田间种群对五种杀虫剂的抗性监测OA
Resistance monitoring of field populations of Tuta absoluta(Lepidoptera:Gelechiidae)in Hebei Region,North China to five insecticides
[目的]为明确河北地区番茄潜叶蛾Tuta absoluta对5种杀虫剂(高效氯氟氰菊酯、茚虫威、乙基多杀菌素、溴虫氟苯双酰胺和环丙氟虫胺)的抗性现状.[方法]于2023-2025年,采用浸叶法测定上述5种杀虫剂对采自河北省张家口市经济开发区、张家口市崇礼区、承德市丰宁满族自治县、承德市平泉市、秦皇岛市青龙满族自治县、唐山市滦州市、廊坊市香河县、廊坊市固安县、保定市容城县、衡水市饶阳县和邯郸市肥乡区大棚或露地番茄植株上的番茄潜叶蛾11个田间种群2龄幼虫48 h的致死中浓度(median lethal concentration,LC50)值,以此评估这11个田间种群对这5种杀虫剂的抗性水平.采用PCR扩增结合基因测序,对番茄潜叶蛾敏感品系幼虫钠离子通道击倒抗性(knock-down resistance,kdr)基因的点突变(M918T,T929I和L1014F)进行检测.[结果]河北省11个地区番茄潜叶蛾种群对高效氯氟氰菊酯、乙基多杀菌素、溴虫氟苯双酰胺和环丙氟虫胺的敏感性较高,对茚虫威产生了不同程度的抗性,其中2023年采集的张家口经济开发区种群和秦皇岛青龙满族自治县种群对茚虫威的抗性最高,达到了高抗水平,抗性倍数分别为60.9和56.3;虽然11个种群对高效氯氟氰菊酯的抗性倍数低(仅为0.4~2.3),但高效氯氟氰菊酯对所有种群(包括相对敏感品系)2龄幼虫48 h内的LC50值均很高,介于178.141~953.106 mg/L,并且相对敏感品系kdr基因M918T,T929I和L1014F 3个位点的突变频率分别为20.0%,25.0%和100.0%.[结论]乙基多杀菌素、溴虫氟苯双酰胺和环丙氟虫胺可作为河北地区防治番茄潜叶蛾的推荐药剂,但建议与其他不同作用机理的杀虫剂或其他防控措施轮换使用,以延缓番茄潜叶蛾抗药性的发展.
[Aim]To clarify the resistance status of Tuta absoluta to five insecticides(lambda-cyhalothrin,indoxacarb,spinetoram,broflanilide and cyproflanilide)in Hebei Region,North China.[Methods]Leaf-dipping method was used to determine the median lethal concentration(LC50)values of the above five insecticides against the 2nd instar larvae of the field-collected populations of T.absoluta during 2023-2025.These populations were sampled from tomato plants in greenhouses or open fields across Hebei Province,including Economic Development Zone of Zhangjiakou,Chongli District of Zhangjiakou City,Fengning Manchu Autonomous County of Chengde City,Pingquan City of Chengde City,Qinglong Manchu Autonomous County of Qinhuangdao,Luanzhou City of Tangshan City,Xianghe County of Langfang City,Gu'an County of Langfang City,Rongcheng County of Baoding City,Raoyang County of Hengshui City and Feixiang District of Handan City.The resistance levels of the 11 field populations to the five insecticides were evaluated based on the LC50 values.Point mutations(M918T,T929I and L1014F)in the sodium channel knock-down resistance(kdr)gene in T.absoluta larvae of the susceptible strain were detected using the PCR amplification followed by gene sequencing.[Results]T.absoluta populations from 11 regions in Hebei Province showed high susceptibility to lambda-cyhalothrin,spinetoram,broflanilide and cyproflanilide,but exhibited varying degrees of resistance to indoxacarb.Notably,the populations collected from Economic Development Zone of Zhangjiakou City and Qinglong Manchu Autonomous County of Qinhuangdao City in 2023 showed the highest resistance to indoxacarb,reaching high-resistance levels with the resistance ratios of 60.9 and 56.3,respectively.Although the resistance ratios of the 11 field populations to lambda-cyhalothrin remained low(only 0.4-2.3),the LC50values of lambda-cyhalothrin against the 2nd instar larvae of all populations(including the relatively susceptible strain)in 48 h were notably high,ranging from 178.141 to 953.106 mg/L.Moreover,the mutation frequencies of the M918T,T929I and L1014F sites in kdr of the relatively susceptible strain were 20.0%,25.0%and 100.0%,respectively.[Conclusion]Spinetoram,broflanilide and cyproflanilide are recommended for controlling T.absoluta in Hebei Region.To delay the development of resistance,these insecticides should be rotated with other insecticides exhibiting different modes of action or integrated with other control measures.
