目的 建立水体中常见病毒实时荧光定量PCR检测方法并评估黄河流域水体中诺如病毒、肠道病毒、甲型肝炎病毒和F型腺病毒等污染情况.方法 首先建立水体中常见病毒的定量PCR检测方法,再采用吸附-洗脱法富集病毒,采用超滤方法浓缩病毒并通过定量PCR技术进行病毒定量检测.结果 在2024年9-10月(丰水期)与2025年3月(枯水期)2个时段采集的22份水样中,诺如病毒与肠道病毒均未检出;27.27%的样本检出甲型肝炎病毒,18.18%的样本中检测出F型腺病毒,检出浓度为1.3~2.1 lgGC/L.结论 黄河流域水体未检出诺如病毒和肠道病毒,个别地点可能受到甲型肝炎病毒和F型腺病毒污染.该结果可为黄河流域水体生物安全评价与控制提供参考.

Objective To establish a real-time quantitative PCR(qPCR)method for detection of common viruses in a water environment,and to assess the severity of contamination by norovirus,enterovirus,hepatitis A virus,and human adenovirus F in the Yellow River basin.Methods A qPCR method for detection of common viruses was established.Viruses were enriched via adsorption-elution,concentrated via ultrafiltration,and quantified by qPCR.Results Twenty-two samples were collected during the high-flow season(September-October 2024)and the low-flow season(March 2025).Neither norovirus nor enterovirus was detected in the samples.Hepatitis A virus was detected in 27.27%of the samples,and human adenovirus F in 18.18%.The concentrations of the detected viruses ranged from 1.3 to 2.1 lgGC/L.Conclusion Norovirus and enterovirus are not detected while contamination by hepatitis A virus and human adenovirus F is observed in some samples.These results may provide a reference for biosafety assessment and management of water resources in the Yellow River basin.
