目的 明确音乐治疗对运动抗抑郁的协同作用并探究其机制.方法 采用慢性束缚应激(CRS)方法构建小鼠抑郁样模型,观察单纯运动及运动联合音乐对抑郁的不同影响,包括使用高架十字迷宫实验和强迫游泳实验观察小鼠的行为学差异;通过小鼠海马组织转录组学测序探究音乐增强运动抗抑郁效果的作用机制;使用免疫荧光评估小鼠脑内小胶质细胞活化情况;使用逆转录定量-聚合酶链式反应(RT-qPCR)评估小鼠系统性炎症水平的变化;使用尼氏染色及高尔基染色分析小鼠神经元损伤及突触可塑性变化.结果 在行为学检测中,与对照组比较,CRS组小鼠出现明显抑郁样行为,运动干预可逆转以上变化,音乐联合运动干预可进一步改善抑郁样行为.海马组织转录组学结果提示音乐可能通过影响神经炎症发挥增强运动抗抑郁效果.免疫荧光、RT-qPCR结果显示,与单纯运动比较,运动联合音乐可显著降低外周炎症细胞因子水平,改善全身系统性炎症;尼氏染色及高尔基染色结果表明,音乐联合运动可更有效逆转抑郁介导的神经元损伤,表现为海马区尼氏体数量和树突棘密度的显著提高.结论 音乐联合运动改善CRS小鼠抑郁样行为,其机制可能与更好地缓解神经炎症,促进尼氏体数量和树突棘密度恢复有关.

Objective To explore the synergistic antidepressant effects of music therapy combined with exercise and to elucidate the underlying mechanisms.Methods A depressive-like mouse model was established using the chronic restraint stress(CRS)paradigm.The effects of exercise alone and exercise combined with music were assessed.Behavioral alterations were evaluated using the elevated plus maze and forced swim tests.Hippocampal transcriptomic sequencing was performed to investigate potential molecular mechanisms by which music enhanced the antidepressant efficacy of exercise.Microglial activation was examined via immunofluorescence staining.Systemic inflammatory changes were quantified using RT-qPCR.Neuronal injury and synaptic plasticity were assessed using Nissl staining and Golgi staining,respectively.Results Behavioral assessments showed that CRS induced distinct depressive-like behaviors,which were reversed by exercise and improved by the combined music-exercise intervention.Transcriptomic analysis suggested that music could enhance the antidepressant effects of exercise by modulating neuroinflammatory pathways.Immunofluorescence and RT-qPCR results indicated that the combined intervention could more significantly reduce levels of peripheral inflammatory cytokines and ameliorate systemic inflammation than exercise alone.Nissl and Golgi staining revealed that music combined with exercise more effectively mitigated depression-related neuronal damage,as evidenced by increased Nissl body abundance and enhanced dendritic spine density in the hippocampus.Conclusion The combination of music and exercise can more effectively improve depressive-like behaviors in CRS mice than exercise alone.This enhanced benefit may be associated with attenuation of neuroinflammation and improved recovery of neuronal integrity and synaptic plasticity.
