目的:探讨加味薏苡附子败酱散对葡聚糖硫酸钠(dextran sulfate sodium,DSS)诱导的溃疡性结肠炎(ulcerative colitis,UC)模型大鼠肠黏膜微循环障碍的改善作用.方法:60只SPF级大鼠随机分为空白组、模型组、美沙拉秦组及加味薏苡附子败酱散低、中、高剂量组,除空白组外各组大鼠予3%DSS造模,成功建立UC模型后,美沙拉秦组及各剂量中药组分别予相应药物灌胃,空白组、模型组予等量生理盐水灌胃.H&E染色观察各组结肠组织病理变化,血液流变仪检测血液流变学参数,ELISA检测大鼠血清P-选择素、TXB2和6-Keto-PGF1α含量,透射电镜观察肠黏膜微血管超微结构,MASSON染色观察微血管胶原纤维阳性面积比率,免疫组化法检测结肠组织VEGF表达、肠黏膜MVD.结果:与空白组相比,模型组结肠病理损伤明显,血液流变学参数、P-选择素、TXB2、VEGF的水平及MVD明显升高(P<0.05),6-Keto-PGF1α水平明显降低(P<0.01),微血管损伤明显,微血管胶原纤维阳性面积比率明显升高(P<0.01).与模型组相比,各给药组结肠病理损伤改善,血液流变学参数、P-选择素、TXB2、VEGF的水平及MVD降低(P<0.05),6-Keto-PGF1α水平明显升高(P<0.01),微血管损伤减轻,微血管胶原纤维阳性面积比率明显降低(P<0.01),中药组中以高剂量组疗效最为明显.结论:加味薏苡附子败酱散可有效改善UC大鼠肠黏膜微循环障碍.

Objective:Explore the improvement effect of the Modified Yiyi Fuzi Baijiang Powder on intestinal mucosal micro-circulatory disturbance in rats with ulcerative colitis(UC)induced by dextran sulfate sodium(DSS).Methods:A total of 60 SPF-grade rats were randomly divided into a blank group,model group,mesalazine group,and low,medium,and high-dose Modified Yiyi Fuzi Baijiang Powder groups.Except for the blank group,rats in all other groups were treated with 3%DSS to establish the UC model.Following successful model establishment,rats in the mesalazine group and each dose of the traditional Chinese medi-cine group received corresponding treatments via gavage,while rats in both the blank and model groups were given an equivalent amount of physiological saline as a control.H&E staining was used to observe the pathological changes of colon tissue in each group.A blood rheology instrument was used to measure hematological parameters.ELISA was employed to detect serum levels of P-selectin,TXB2,and 6-Keto-PGF1α in the rats.Transmission electron microscopy was utilized to examine the ultrastructure of intestinal microvessels,while Masson's trichrome staining assessed the ratio of microvascular collagen fiber-positive area.Immu-nohistochemical methods were used to detect the levels of VEGF in colonic tissue and to measure MVD in the intestinal mucosa.Result:Compared to the blank group,the model group showed significant pathological damage to the colon,with significantly in-creased levels of blood rheological parameters,P-selectin,TXB2,VEGF,and MVD(P<0.05).The expression level of 6-Keto-PGF1α was significantly decreased(P<0.01),microvascular damage was significant,and the ratio of microvascular collagen fiber-positive area was significantly increased(P<0.01).Compared to the model group,each treatment group showed improvement in colonic pathological damage,with decreased levels of blood rheological parameters,P-selectin,TXB2,VEGF,and MVD(P<0.05).The expression level of 6-Keto-PGF1α was increased(P<0.01),microvascular damage was alleviated,and the ratio of mi-crovascular collagen fiber-positive area was significantly reduced(P<0.01).Among the traditional Chinese medicine groups,the high-dose group demonstrated the most significant efficacy.Conclusion:Modified Yiyi Fuzi Baijiang Powder can effectively im-prove intestinal mucosal microcirculatory disturbance in rats with UC.
