辣椒主要病害广谱生防木霉菌的筛选与评价及鉴定OA
Screening,Evaluation and Identification of Trichoderma,a Broad-spectrum Biocontrol Fungus,Against Major Diseases in Chili Pepper
[目的]分离筛选对辣椒枯萎病、疫病、白绢病、炭疽病等具有广谱生防作用的木霉菌,为辣椒主要病害可持续防控提供生防资源.[方法]采集辣椒不同生长期的根际土壤,利用梯度稀释法和平板对峙法分离和筛选对 4种辣椒常发性病害病原菌拮抗效果俱佳的菌株,通过菌落直径法测定其发酵液抑菌活性,通过盆栽及离体果实防效试验评价其生防作用,并结合形态学和分子生物学对菌株进行鉴定.[结果]从土样中共分离到木霉菌 10株,其中,菌株 YDMM-07对辣椒枯萎病病原菌(Fusarium oxysporum)、白绢病病原菌(Athelia rolfsii)、疫病病原菌(Phytophthora capsici)、炭疽病病原菌(Colletotrichum capsici)的抑制率均最高,分别达 83.71%、81.79%、93.24%、85.51%;且其无菌发酵液对 4种病原菌的抑制效果分别为 49.63%、31.82%、58.30%、63.22%.菌株 YDMM-07含菌发酵产物对辣椒枯萎病、疫病、白绢病的盆栽防效分别为66.98%、71.00%、62.98%,其无菌发酵滤液对炭疽病离体果实防效为 52.39%.经形态学和分子生物学鉴定,菌株YDMM-07为长枝木霉(Trichoderma longibrachiatum).[结论]长枝木霉 YDMM-07对辣椒枯萎病、疫病、白绢病、炭疽病4种常发性病害有较好的防治效果,可作为辣椒病害生防菌剂资源进行开发.
[Objective]The Trichoderma strains with a broad-spectrum biocontrol effect against 4 major diseases of Fusarium oxysporum,Phytophthora capsici,Athelia rolfsii and Colletotrichum capsici in chili pepper were isolated and screened to provide biocontrol resources for sustainable prevention and control of major diseases in chili peppers.[Method]The Trichoderma strains with an optimal antagonistic effect against 4 common diseases were isolated and screened from the rhizosphere soil of chili peppers at different growth stages by the gradient dilution method and plate confrontation method.The inhibition activity of the fermentation liquid of Trichoderma strains was determined by the colony diameter method.Meanwhile,the biocontrol effect was evaluated by the pot and in vitro pepper prevention and control experiments.The screened strains were identified in morphology and molecular biology.[Result]10 Trichoderma strains were isolated from the soil samples.The inhibitory rate of strain YDMM-07 against F.oxysporum,A.rolfsii,P.capsici and C.capsici was 83.71%,81.79%,93.24%and 85.51%respectively.The inhibitory rate of strain YDMM-07 sterile fermentation broth against F.oxysporum,A.rolfsii,P.capsici and C.capsici was 49.63%,31.82%,58.30%and 63.22%,separately.The prevention and control rate of strain YDMM-07 fermentation broth containing mycelia against F.oxysporum,P.capsici and A.rolfsii was 66.98%,71.00%and 62.98%in the pot experiment respectively,and the control rate of the sterile fermentation broth against C.capsici of in vitro pepper reached 52.39%.Strain YDMM-07 was identified as Trichoderma longibrachiatum according to its morphology and molecular biology.[Conclusion]T.longibrachiatum YDMM-07 strain with a good prevention and control effect against 4 major diseases of F.oxysporum,P.capsici,A.rolfsii and C.capsici can be used in development of antibiological agents against diseases in chili pepper.
