基于BSA-seq技术的饲料油菜植株粗蛋白含量QTL定位研究OA
QTL Mapping of Crude Protein Content in Forage Rape Plants Based on BSA-seq Technology
[目的]解析饲料油菜植株粗蛋白性状调控遗传位点及关键基因,为饲料油菜植株粗蛋白性状改良和优质饲料油菜品种选育提供参考.[方法]利用 BSA-seq技术,对植株极端粗蛋白差异的饲料油菜21SL-239、21SL-756亲本和极端差异的 F2 分离群体 DNA混池测序,并进行关联分析.[结果]在油菜 C08染色体50.46~53.02 Mb区间共鉴定37个与粗蛋白含量性状显著关联区,通过功能注释筛选出ZS11C08G057720、ZS11C08G059050、ZS11C08G058760等 38个与蛋白质合成、氨基酸供应、蛋白质降解相关的功能基因,ZS11C-08G058820、ZS11C08G057600、ZS11C08G061500、ZS11C08G061580、ZS11C08G061590等 97个与碳水化合物转运和代谢、能量生产和转换、转录调控相关的功能基因,ZS11C08G058790、ZS11C08G061450、ZS11C08G059590等104个功能未知基因.[结论]定位 1个 QTL位点与饲料油菜植株粗蛋白含量相关,并筛选出 239个可能与粗蛋白含量相关的候选基因.
[Objective]The genetic loci and key genes regulating crude protein trait of forage rapeseed plants were analyzed to provide a reference for improvement of crude protein trait of forage rapeseed plants and breeding of high-quality forage rapeseed varieties.[Method]The BSA-seq technology was used to sequence and conduct association analysis on DNA mixed pool samples of parental lines 21SL-239 and 21SL-756 with extreme crude protein differences and F2 segregating population with extreme differences.[Result]A total of 37 regions significantly associated with crude protein content trait were identified within the 50.46-53.02 Mb interval on chromosome C08 of rapeseed.38 functional genes(ZS11C08G057720,ZS11C08G059050,ZS11C08G058760,etc.)related to protein synthesis,amino acid supply,protein degradation,97 functional genes(ZS11C08G058820,ZS11C08G057600,ZS11C08G061500,ZS11C08G061580,ZS11C08G061590,etc.)related to carbohydrate transport and metabolism,energy production and conversion,transcription regulation,etc.and 104 unknown function genes(ZS11C08G058790,ZS11C08G061450,ZS11C08G059590,etc.)were screened by a functional annotation technology.[Conclusion]One QTL locus related to crude protein content of forage rapeseed plants is located,and 239 candidate genes potentially related to crude protein content are screened.
