注意力缺陷多动障碍动物模型的中西医临床病证特点评价分析OA
Evaluation and Analysis of Animal Models of Attention Deficit Hyperactivity Disorder Based on Characteristics of Clinical Symptoms of Traditional Chinese and Western Medicine
目的:基于注意力缺陷多动障碍(attention deficit and hyperactivity disorder,ADHD)的中西医临床病证特点,对现有 ADHD 动物模型进行评价分析,为推进 ADHD 的研究发展提供理论基础.方法:计算机检索数据库,总结归纳国内外现有的 ADHD 动物造模方法,通过查阅 ADHD 及其动物模型的相关文献,整理概述其中西医病因病机、诊断标准,基于中西医临床病证特点建立动物模型评价体系,依据赋值法量化中西医临床吻合度,对ADHD 动物模型的特点进行综合评价分析并阐明其优缺点.结果:共纳入文献60 篇,其中包含35 种 ADHD 动物模型,并将其分为三个类别:遗传模型、化学/环境诱导模型和多因素模型.发现目前ADHD 动物模型具有以下特点:遗传模型与化学/环境诱导模型相比,造模方法更多样且中西医临床吻合度更具优势;中西医临床吻合度较高的是原发性高血压大鼠(SHR)模型,其具有重现性好和成本低的优点;西医临床吻合度普遍高于中医临床吻合度;缺乏多因素及中医病证结合的 ADHD 动物模型.结论:现有的模型大多应用单因素造模方法,中医证候的体现较少,未来应积极开发中西医临床病证高度吻合的 ADHD 动物模型.
Objective:This article aims to evaluate and analyze existing animal models of attention deficit hyperac-tivity disorder(ADHD)based on the clinical symptom characteristics of ADHD in traditional Chinese medicine(TCM)and Western medicine,so as to provide a theoretical basis for the advancement of ADHD research and development.Methods:Through computer database retrieval,the existing ADHD animal modeling methods in China and abroad were summarized,and by reviewing the relevant literature on ADHD and its animal models,the etiology,pathogenesis,and di-agnostic criteria of TCM and Western medicine were compiled and summarized.According to the clinical symptom char-acteristics in TCM and Western medicine,an evaluation system for animal models was established,and the model's clini-cal fit in TCM and Western medicine was systematically quantified using the method of assignment so that the characteris-tics of the ADHD animal model could be visualized and comprehensively analyzed,and its advantages and disadvantages could be elucidated.Results:A total of 60 articles were included,containing 35 types of ADHD animal models,which were classified into three categories:genetic models,chemical/environmental induction models,and multifactor models.In the evaluation and analysis of the above ADHD animal models,it is found that compared with chemical/environmental induction models,genetic models have a greater variety of modeling methods and a more advantageous clinical fit in TCM and Western medicine.The spontaneously hypertensive rat(SHR)model has the highest clinical fit in TCM and West-ern medicine,with the advantages of good reproducibility and low cost.It is also found that the clinical fit in Western medicine is generally higher than the clinical fit in TCM,and there is a lack of ADHD animal models with multiple factors and symptoms in TCM.Conclusions:Most existing models use single-factor modeling methods,with less symptom repre-sentation in TCM.In the future,it is necessary to actively develop ADHD animal models highly compatible with the clini-cal symptom characteristics in TCM and Western medicine.
