目的 探究温阳活血方对子宫腺肌病小鼠胞饮突的改善情况及作用机制.方法 使用他莫昔芬法建立子宫腺肌病小鼠模型,设立正常组、模型组、促性腺激素释放激素激动剂(GnRH-α)组及温阳活血方低、中和高剂量组.各组药物干预 8 周.使用扫描电镜观察各组胞饮突数量,蛋白免疫印迹法、免疫组化法观察各组胞饮突生成通路叉头框蛋白A2(FOXA2)/白血病抑制因子(LIF)/同源框蛋白A10(HOXA10)的表达变化,q-PCR法检测各组小鼠子宫内膜FOXA2 和HOXA10 的mRNA表达.结果 与正常组比较,子宫腺肌病模型小鼠的子宫内膜未观察到明显的胞饮突(P<0.05);与正常组比较,模型组小鼠子宫内膜中FOXA2、LIF和HOXA10 蛋白表达均降低(P<0.05),FOXA2 和HOXA10 的mRNA表达水平明显降低(P<0.01).与模型组比较,温阳活血方高、中和低剂量均能够增加子宫腺肌病小鼠子宫内膜的胞饮突数量(P<0.05).与模型组比较,温阳活血方高剂量组能够明显上调FOXA2、LIF和HOXA10 蛋白表达水平(P<0.05),并上调FOXA2 和HOXA10 的mRNA表达水平(P<0.01).温阳活血方高剂量组与GnRH-α组比较,作用相近(P>0.05).结论 温阳活血方能够激活FOXA2/LIF/HOXA10 通路,增加子宫腺肌病小鼠子宫内膜胞饮突的数量,可为临床改善子宫腺肌病不孕患者容受性作用提供机制参考.

Objective To investigate the effect of Wenyang Huoxue Formula on improving pinopodes in adenomyosis mice and its underlying mechanism.Methods A mouse model of adenomyosis was established using the tamoxifen method.Mice were divided into normal control group,model group,GnRH-α(gonadotropin-releasing hormone agonist)group,and low-,medium-,and high-dose Wenyang Huoxue Formula groups.Each group received the corresponding intervention for 8 weeks.The number of pinopodes was observed by scanning electron microscopy.The expression levels of Forkhead box A2(FOXA2),Leukemia Inhibitory Factor(LIF),and homeobox A10(HOXA10)in the pinopode formation pathway were detected by Western Blot and immunohistochemistry.The mRNA expression levels of FOXA2 and HOXA10 in the mouse endometrium were measured by q-PCR.Results Compared with the normal control group,no obvious pinopodes were observed in the endometrium of adenomyosis model mice(P<0.05).The protein expression levels of FOXA2,LIF,and HOXA10 in the model group were significantly lower than those in the normal control group(P<0.05),and the mRNA expression levels of FOXA2 and HOXA10 were also significantly decreased(P<0.01).Compared with the model group,all doses(low,medium,and high)of Wenyang Huoxue Formula significantly increased the number of pinopodes in the endometrium of adenomyosis mice(P<0.05).The high-dose Wenyang Huoxue Formula group significantly upregulated the protein expression levels of FOXA2,LIF,and HOXA10(P<0.05)as well as the mRNA expression levels of FOXA2 and HOXA10(P<0.01)compared with the model group.Furthermore,the effect of the high-dose Wenyang Huoxue Formula was similar to that of GnRH-α(P>0.05).Conclusion Wenyang Huoxue Formula can activate the FOXA2/LIF/HOXA10 pathway to increase the number of pinopodes in the endometrium of adenomyosis mice,providing a mechanistic reference for improving endometrial receptivity in patients with adenomyosis-associated infertility.
