穴位注射磁性水凝胶对CCI大鼠脊髓背角JAK1/STAT3表达的影响OA
Effect of Acupoint Injection of Magnetic Hydrogel on JAK1/STAT3 Expression in the Spinal Dorsal Horn of CCI Rats
目的:本研究旨在制备一种负载磁性纳米粒子(Fe3O4)的壳聚糖/β-甘油磷酸钠(CS/GP)温敏型磁性水凝胶,并评估其对SD大鼠慢性压缩性坐骨神经痛(CCI)模型的治疗效果,以探索其作为穴位埋线替代材料的可行性.方法:通过CS/GP/Fe3O4水凝胶体外实验来观察其成胶性及细胞相容性;CCI造模前和模型后第1、3、5、7、14天检测大鼠足底机械痛阈值.治疗组于患侧足三里穴(ST36)注射CS/GP/Fe3O4水凝胶,对照组则在相同穴位植入羊肠线.术后第14天取脊髓组织,采用Western blot检测脊髓背角中JAK1和STAT3蛋白的表达水平,以研究其镇痛机制.结果:温敏性实验表明CS/GP/Fe3O4水凝胶常温下呈液态,溶液状态有利于其可注射,37 ℃左右固化呈胶状,具有良好的温敏性;细胞相容性实验表明水凝胶与成纤维细胞具有良好的生物相容性.行为学可观察到注射CS/GP/Fe3O4水凝胶后能够提高CCI大鼠机械痛阈值.Western blot结果示CS/GP/Fe3O4水凝胶能够抑制CCI模型后脊髓背角STAT3和JAK1的表达,且抑制效果优于羊肠线组.结论:CS/GP/Fe3O4磁性温敏水凝胶具有良好的生物相容性、可注射性和镇痛效果,有望作为穴位埋线治疗坐骨神经痛的一种新型替代材料.
Objective This study aimed to prepare a thermosensitive magnetic hydrogel loaded with magnetic nanoparticles(Fe3O4)based on chitosan/β-glycerophosphate(CS/GP),and to evaluate its therapeutic effect on a chronic constriction injury(CCI)model of sciatic nerve pain in SD rats,exploring its feasibility as an alternative material for acupoint catgut embedding. Methods The gelation properties and cytocompatibility of the Fe3O4-loaded chitosan/glycerophosphate(CS/GP/Fe3O4)hydrogel were observed through in vitro experiments.Mechanical paw withdrawal thresholds were measured before CCI modeling and on days 1,3,5,7,and 14 postmodeling.The treatment group received injections of CS/GP/Fe3O4 hydrogel at the Zusanli(ST36)acupoint on the affected side,while the control group received catgut embedding at the same acupoint.Spinal cord tissue was collected on day 14 posttreatment,and the Western blot was used to detect the expression levels of janus kinase 1(JAK1)and signal transducer and activators of transcription 3(STAT3)proteins in the spinal dorsal horn to investigate the analgesic mechanism. Results Thermosensitivity tests showed that the CS/GP/Fe3O4 hydrogel was liquid at room temperature(suitable for injection)and solidified into a gel at around 37℃,demonstrating good thermosensitivity.Cytocompatibility tests indicated good biocompatibility of the hydrogel with fibroblasts.Behavioral tests showed that injection of the CS/GP/Fe3O4 hydrogel could increase the mechanical pain threshold in CCI rats.Western blot results showed that the CS/GP/Fe3O4 hydrogel could inhibit the expression of STAT3 and JAK1 in the spinal dorsal horn after CCI modeling,and the inhibitory effect was superior to that of the catgut embedding group. Conclusion The CS/GP/Fe3O4 magnetic thermosensitive hydrogel exhibits good biocompatibility,injectability,and analgesic effects,showing promise as a novel alternative material for acupoint catgut embedding therapy in treating sciatic nerve pain.
