复方赤石脂禹余粮散对小鼠止泻作用机制研究OA

探究复方赤石脂禹余粮散对小鼠止泻的作用机制.用ICP-OES检测主要元素含量,吸湿率法测定吸湿度.将70只ICR小鼠随机分为正常、模型、赤石脂禹余粮汤、赤石脂散、禹余粮散、赤石脂禹余粮散和阳性对照组.灌胃蓖麻油制备腹泻模型,测量肠道伊文思蓝推进率,观察小肠和盲肠肠壁组织病理改变,检测尿素氮、锌离子、肿瘤坏死因子-α(TNF-α)含量和Na+-K+ATP酶活力,免疫组化检测白细胞介素-6(IL-6)和白细胞介素-1β(IL-1β)蛋白表达,16S rRNA测序检测盲肠内容物.结果显示,赤石脂禹余粮散的主要元素为Fe和Al,其次包括Ca、Mn、Mg、Zn等,可显著抑制腹泻小鼠的肠道伊文思蓝推进率,降低肠壁细胞损伤及血清TNF-α含量,提高肠壁细胞Na+-K+ATP酶活力和血液中锌离子浓度,降低肠壁组织中IL-6和IL-1β的蛋白表达,降低葡萄球菌属(Staphylococcus)和幽门螺旋杆菌属(Helicobacter)等相对丰度而增加乳杆菌属(Lactobacillus)、毛螺菌属(Faecalibacterium)和双歧杆菌属(Bifidobacterium)等有益菌丰度.因此,赤石脂禹余粮散通过补充微量元素,吸附炎症渗出及调节肠道菌群平衡等机制发挥止泻作用.

The study explores the mechanism of anti-diarrheal effect of Compound Halloysitum Rubrum and Limonitum Powder in mice.The main element content was detected by ICP-OES,and the hygroscopicity was determined by the hygroscopic rate method.A total of 70 ICR mice were randomly divided into normal,model,Halloysitum Rubrum and Limonitum Decoction,Halloysitum Rubrum Powder,Limonitum Powder,Halloysitum Rubrum and Limonitum Powder,and positive control groups.Diarrhea models were prepared by gavage with castor oil,and the intestinal Evan's blue propulsion rate was measured.The pathological changes in the small and cecal intestinal wall tissues were observed,and the urea nitrogen,zinc ions,tumor necrosis factor-α(TNF-α)content,and Na+-K+ATPase activity were detected.Immunohistochemistry was used to detect the protein expression of interleukin-6(IL-6)and interleukin-1β(IL-1β),and 16S rRNA sequencing was performed on cecal contents.The results showed that the main elements of Halloysitum Rubrum and Limonitum Powder are Fe and Al,followed by Ca,Mn,Mg,Zn,etc.It significantly inhibited the intestinal Evan's blue propulsion rate in diarrheal mice,reduced intestinal wall cell damage and serum TNF-α content,increased intestinal wall cell Na+-K+ATPase activity and blood zinc ion concentration,and decreased the protein expression of IL-6 and IL-1β in intestinal wall tissues.It also reduced the relative abundance of Staphylococcus and Helicobacter while increasing the abundance of beneficial bacteria such as Lactobacillus,Faecalibacterium,and Bifidobacterium.Therefore,Halloysitum Rubrum and Limonitum Powder exerts its anti-diarrheal effect by supplementing trace elements,adsorbing inflammatory exudates,and regulating the balance of intestinal flora.
