重组表达S100A9蛋白枯草芽孢杆菌对哮喘小鼠的干预作用OA

为阐明S100A9重组蛋白对哮喘疾病的治疗作用,本研究利用OVA诱导的哮喘小鼠模型,从分子与病理水平探究了重组表达S100A9枯草芽孢杆菌对哮喘小鼠的治疗作用和最佳剂量.结果表明:与哮喘及空载对照组比较,低剂量重组菌(1.02×107 CFU/mL)干预组小鼠的喘息、弓背等症状显著减轻,体重增加,肺系数减小;与低剂量重组菌处理组比较,中剂量(1.18×108 CFU/mL)和高剂量处理组(1.25×109 CFU/mL)治疗效果不及低剂量组;病理切片结果也显示低剂量处理组小鼠气道炎症细胞浸润明显减少,小鼠支气管内壁增厚程度减轻(P<0.05);肺组织促炎因子I L-5、I L-6、I L-13的mRNA表达水平显著降低,气道粘液蛋白Muc5ac、Muc5b的mRNA表达水平也显著下降(P<0.05);小鼠血清中细胞因子IL-6、TNF-α的含量显著降低;肺组织SOD和CAT酶含量增加,MDA指标有明显增加;肺泡灌洗液中嗜酸性粒细胞等炎症细胞数显著降低(P<0.05).上述结果表明低剂量的S100A9重组枯草芽孢菌对哮喘干预具有一定的治疗效果.

To investigate the therapeutic effect of S100A9 recombinant Bacillus subtilis on asthma,the effects and optimal dosage of S100A9 recombinant Bacillus subtilis was explored using the OVA-induced asthmatic mouse from the molecular expression and pathological levels.Compared with the groups of asthma and vector control,the low-dose recombinant bacteria group(1.02×107 CFU/mL)showed a significant reduction in actions of wheezing and arching back,an increase in body weight,and a reduction in lung coefficients;compared with the low-dose,the effects were weaker in the medium-dose-treated group(1.18×108 CFU/mL)and the high-dose-treated group(1.25×109 CFU/mL);pathological sections showed a significant reduction of inflammatory cells infiltration in the airways,and a reduction of thickness of the bronchial tubes(P<0.05);and the expressions of IL-5,IL-6,and IL-13 were significantly decreased in the lung tissues;the mRNA levels of Muc5ac and Muc5b were also significantly reduced(P<0.05);the levels of IL-6 and TNF-α in the serum of mice in the low-dose recombinant bacteria-treated group was significantly reduced;the content of SOD and CAT enzymes in the lung tissues were increased,and there was a significant recovery of the MDA indexes;the number of eosinophils in the alveolar lavage fluid was significantly reduced(P<0.05).The above results indicated that low-dose S100A9 recombinant Bacillus subtilis had a better treating effects on asthma.
