长柔毛野豌豆(Vicia villosa)是野豌豆属一年生植物,喜欢生长在湿润且富有机质的生境中.近年来豆科植物叶绿体基因组研究已取得阶段性进展.本研究基于NCBI长柔毛野豌豆全基因组重测序数据,对其叶绿体基因组展开组装与分析.结果显示:长柔毛野豌豆叶绿体基因组全长为124228 bp,GC含量34.63%,同时缺少反向重复IR区,不具备典型的四分体结构,属于缺乏反向重复序列的物种.长柔毛野豌豆叶绿体基因组涵盖110个基因,包括76个CDS、4个rRNA基因及30个tRNA基因.密码子分析显示,其叶绿体基因组密码子更偏好使用A/T碱基.重复序列分析检测到65个SSRs,这些位点多以单核苷酸重复形式存在,且长重复序列主要以正向重复为主.另外,与近缘物种相比,它们的叶绿体基因组间变异整体较小,但在ycf4、rps11、rpoA、trnL-UAG~trnF-GAA、trnI-CAU、trnL-CAA、trnH-GUG~trnN-GUU和ndhB区域存在明显差别.系统发育分析显示,长柔毛野豌豆与广布野豌豆亲缘关系较近,表明二者在进化历程中可能拥有共同的祖先.本研究为野豌豆属植物遗传多样性分析和野生资源开发利用等工作提供基础的数据支持.

Vicia villosa belongs to the Vicia genus of the Fabaceae family and is an annual herb that prefers to grow in moist and organic-rich habitats.Although the chloroplast genome study of Fabaceae has gradually progressed in recent years,the chloroplast genome of Vicia villosa,a member of the IRLC(inverted repeat lacking clade)taxon,has not yet been assembled.This study is based on NCBI V.villosa whole genome resequencing data and assembled and analyzed its chloroplast genome.These results showed that the full length of the V.villosa chloroplast genome is 124228 bp,with 34.63%GC content,and it lacks the inverted repeat IR region,does not have the typical tetrad structure,and belongs to the species lacking in inverted repeat sequence.This chloroplast genome has 110 genes,including 76 CDS,4 rRNA genes and 30 tRNA genes.Codon analysis showed a preference for A/T bases for its chloroplast genome codons.Repeat sequence analyses detected 65 SSRs,and these loci were mostly in the form of mononucleotide repeats,and in the case of long repeat sequences mainly forward repeats were dominant.In addition,the overall variation among their chloroplast genomes was small compared to their close relatives,but significant differences existed in the ycf4,rps11,rpoA,trnL-UAG~trnF-GAA,trnI-CAU,trnL-CAA,trnH-GUG~trnN-GUU,and ndhB regions.Phylogenetic analyses showed that Vicia villosa and Vicia cracca are relatively close to each other,indicating that they may share a common ancestor during the evolutionary process.This study provides some basic data support for the genetic diversity analysis of Vicia and the development and utilization of wild resources.
