针刺联合姜黄素调控Shh/Gli1通路对慢性萎缩性胃炎大鼠胃粘膜损伤的影响OA
Effects of Acupuncture Combined with Curcumin on Gastric Mucosal Injury in Rats with CAG via Regulation of Shh/Gli1 Pathway
目的:探究针刺联合姜黄素(Cur)调控音猬因子(Shh)/胶质瘤相关癌基因同源物(Gli1)通路对慢性萎缩性胃炎(CAG)大鼠胃粘膜损伤的影响.方法:建立CAG大鼠模型,将建模成功的大鼠随机分为CAG组、针刺组和Cur组、Cur+针刺组和Cur+针刺+Shh/Gli1通路抑制剂cyclopamine(Cur+针刺+cy-clopamine组),每组15只.另外随机选取15只正常大鼠为NC组.检测大鼠血清炎性相关因子和胃功能相关指标表达;检测大鼠胃组织病理学变化;观察胃粘膜细胞超微结构;检测大鼠胃粘膜细胞凋亡变化;免疫组化检测大鼠胃组织凋亡相关蛋白表达;Western blot检测大鼠胃组织Shh/Gli1通路相关蛋白表达.结果:与NC组比较,CAG组大鼠胃粘膜变薄,腺体萎缩,排列疏松紊乱,大量炎性细胞浸润,线粒体结构异常,嵴稀疏且断裂,IL-6、IL-8、TNF-α、胃动素(MTL)、细胞凋亡率和Bax表达升高,胃泌素(GAS)、生长抑素(SS)、胃蛋白酶(PP)、Bcl-2、Shh和Gli1表达降低,差异具有统计学意义(P<0.05);与CAG组比较,针刺组、Cur组和Cur+针刺组大鼠胃粘膜损伤改善,炎性细胞浸润减少,线粒体结构恢复,IL-6、IL-8、TNF-α、MTL、细胞凋亡率和 Bax 表达降低,GAS、SS、PP、Bcl-2、Shh 和 Gli1 表达升高,差异具有统计学意义(P<0.05);与针刺组、Cur组比较,Cur+针刺组大鼠胃粘膜损伤、炎性细胞浸润和线粒体结构损伤较轻,IL-6、IL-8、TNF-α、MTL、细胞凋亡率和Bax表达低,GAS、SS、PP、Bcl-2、Shh和Gli1表达高,差异具有统计学意义(P<0.05);与Cur+针刺组比较,Cur+针刺+cyclopamine组大鼠胃粘膜损伤和炎性细胞浸润加重,线粒体肿胀、结构异常,IL-6、IL-8、TNF-α、MTL、细胞凋亡率和Bax表达升高,GAS、SS、PP、Bcl-2、Shh和Gli1表达降低,差异具有统计学意义(P<0.05).结论:针刺联合姜黄素能够改善CAG大鼠胃功能,对胃粘膜损伤起到保护作用,可能与激活Shh/Gli1通路密切相关.
Objective:To investigate the effects of acupuncture combined with curcumin(Cur)on gastric mucosal injury in rats with chronic atrophic gastritis(CAG)through regulation of the Sonic hedgehog/glioma-associated oncogene homolog 1(Shh/Gli1)pathway.Methods:The rat model of CAG was established,and the successfully modeled rats were randomly divided into the CAG group,the acupuncture group,the Cur group,the Cur+acupuncture group,and the Cur+acupuncture+cyclopamine(Shh/Gli1 pathway inhibitor)group,with 15 rats in each group.In addition,15 normal rats were randomly selected as the NC group.The expression levels of serum inflammation-related factors and gastric function-related indicators were measured.Histopathological changes in gastric tissues were examined.The ultrastructure of gastric mucosal cells was observed.Apoptosis of gastric mucosal cells was assessed.Immunohistochemistry was used to detect the expression of apoptosis-related proteins in gastric tissues,and Western blot was performed to determine the expression of Shh/Gli1 pathway-related proteins in gastric tissues.Results:Compared with the NC group,rats in the CAG group showed thinning of the gastric mucosa,glandular atrophy,loose and disordered glandular arrangement,marked inflammatory cell infiltration,abnormal mitochondrial structure,and sparse and fractured cristae;in addition,the levels of IL-6,IL-8,TNF-α,motilin(MTL),apoptosis rate,and Bax were significantly increased,whereas the levels of gastrin(GAS),somatostatin(SS),pepsinogen(PP),Bel-2,Shh,and Gli1 were significantly decreased(P<0.05).Compared with the CAG group,rats in the acupuncture group,the Cur group,and the Cur+acupuncture group showed improved gastric mucosal injury,reduced inflammatory cell infiltration,and restored mitochondrial structure;moreover,the levels of IL-6,IL-8,TNF-α,MTL,apoptosis rate,and Bax were significantly decreased,whereas the levels of GAS,SS,PP,Bel-2,Shh,and Gli1 were significantly increased(P<0.05).Compared with the acupuncture group and the Cur group,rats in the Cur+acupuncture group exhibited milder gastric mucosal injury,less inflammatory cell infiltration,and less mitochondrial structural damage;the levels of IL-6,IL-8,TNF-α,MTL,apoptosis rate,and Bax were significantly lower,whereas the levels of GAS,SS,PP,Bcl-2,Shh,and Gli1 were significantly higher(P<0.05).Compared with the Cur+acupuncture group,rats in the Cur+acupuncture+cyclopamine group showed aggravated gastric mucosal injury and inflammatory cell infiltration,mitochondrial swelling,and abnormal mitochondrial structure;the levels of IL-6,IL-8,TNF-α,MTL,apoptosis rate,and Bax were significantly increased,whereas the levels of GAS,SS,PP,Bcl-2,Shh,and Gli1 were significantly decreased(P<0.05).Conclusion:Acupuncture combined with curcumin can improve gastric function in rats with CAG and exert a protective effect against gastric mucosal injury,which may be closely related to activation of the Shh/Gli1 pathway.
