为建立巴戟天种质复壮组织培养快繁技术体系,以肇庆主栽品种"黑穗仔"嫩茎尖为外植体,研究MS和N6培养基及不同浓度6-BA、IBA组合对其诱导、分化、增殖和生根的影响,筛选各发育阶段的最适培养基.结果表明,MS培养基诱导效果最佳,初代芽诱导培养基为MS+IBA 0.10 mg/L+6-BA 1.50 mg/L;继代增殖培养基为MS+6-BA 0.5~1.00 mg/L+IBA 0.10 mg/L,不定芽成活率为95.56%~98.23%,分蘖数为6.97~7.66个;壮苗培养基为MS+6-BA0.30 mg/L+IBA 0.10 mg/L;生根培养基为MS+1.00 g/L活性炭+1.00 mg/L IBA,生根率为95.75%,生根系数为11.17.种质复壮组培苗原瓶炼苗15 d后,移栽至泥炭土或泥炭土+河沙基质,70 d成活率达98.33%,生长健壮.

In order to establish a tissue culture rapid propagation technology system for the rejuve-nation of the medicinal plant Morinda officinalis,using the young shoot tips of the main cultivated variety Heisuizai in Zhaoqing as the explants,the effects of MS and N6 culture media and different concentrations of 6-BA and IBA combinations on their induction,differentiation,proliferation and rooting were studied.The most suitable culture media at each developmental stage were screened.The results showed that the MS medium had the best induction effect.The initial generation bud induction medium was MS+IBA 0.10 mg/L+6-BA 1.50 mg/L.The secondary generation proliferation medium was MS+6-BA 0.5-1.00 mg/L+IBA 0.10 mg/L.The survival rate of adventitious buds was 95.56%-98.23%,and the number oftillers was 6.97-7.66.The vigorous seedling culture medium was MS+6-BA 0.30 mg/L+IBA 0.10 mg/L.The rooting culture medium was MS+1.00 g/L activated carbon+1.00 mg/L IBA.The rooting rate was 95.75%,and the rooting coefficient was 11.17.After the o-riginal bottle of the tissue culture seedlings was acclimatized for 15 days,which were transplanted to peat soil or peat soil+river sand substrate.The survival rate reached 98.33%after 70 days and the growth was vigorous.
