为破解柔毛淫羊藿野生资源枯竭与种子深度休眠制约人工栽培的难题,采用低温层积、变温层积、200 mg/L赤霉素(GA3)、10 mg/L氟啶酮(FL)及其复合处理进行破眠,系统观察了 90 d内种胚长、胚宽及胚率的动态变化,并统计发芽率与发芽势.系统分析柔毛淫羊藿种子休眠特性与破眠方法,结果表明,柔毛淫羊藿种子为形态-生理复合休眠类型;胚发育动态显示,200 mg/L GA3处理可有效促进胚的形态后熟,胚率达92.66%;10 mg/L FL处理虽在早期(30 d)促进种胚由球形向心形转变,但其对胚形态的促进效果弱于GA3处理,然而在破除生理休眠、促进萌发方面作用显著,发芽率最高达69.22%.GA3与FL复合处理未产生协同增效.柔毛淫羊藿种子破眠存在形态后熟与生理解锁两条相对独立的通路,其中,GA3主导胚形态建成,而FL则高效解除生理休眠.采用10 mg/L FL浸泡6 h,继以4 ℃黑暗层积90 d,是破除种子休眠、实现高效萌发的优选方案.

To solve the problems of depletion of wild resources of Epimedium pubescens and the deep dormancy of its seeds,which hindered artificial cultivation,various treatments such as low-tempera-ture stratification,variable-temperature stratification,200 mg/L gibberellin(GA3),10 mg/L fluri-done(FL)and their combinations were used to break the dormancy.The dynamic changes of seed em-bryo length,width and germination rate within 90 days were systematically observed,and the germi-nation rate and germination potential were statistically analyzed.The dormancy characteristics of Epimedium pubescens seeds and the breaking dormancy methods were systematically analyzed.The results showed that the seeds of Epimedium pubescens were of a morphological-physiological com-bined dormancy type.The dynamic development of the embryo showed that the 200 mg/L GA3 treat-ment could effectively promote the morphological ripening of the embryo,with an embryo rate of 92.66%;although the 10 mg/L FL treatment promoted the transformation of the seed embryo from spherical to heart-shaped in the early stage(30 days),of which promoting effect on the embryo mor-phology was weaker than that of the GA3 treatment,but it had a significant effect in breaking physio-logical dormancy and promoting germination,with the highest germination rate of 69.22%.The GA3 and FL combined treatment did not show a synergistic effect.The breaking dormancy of Epimedium pubescens seeds existed through 2 relatively independent pathways:morphological ripening and physi-ological unlocking.Among them,GA3 dominated the embryo morphology formation,while FL effi-ciently unlocked the physiological dormancy.Using 10 mg/L FL immersion for 6 hours followed by 4℃ dark stratification for 90 days was the preferred scheme for breaking the seed dormancy and achieving efficient germination.
