A preliminary study on the anti-Candida albicans activity of duloxetine hydrochloride

目的 通过体外实验,系统性评估盐酸度洛西汀对白念珠菌的影响.方法 微量肉汤稀释法检测盐酸度洛西汀抗白念珠菌生长的最低抑菌浓度和最低杀菌浓度;通过时间-杀菌曲线考察盐酸度洛西汀抗白念珠菌的时间杀菌趋势;利用倒置显微镜观察不同浓度盐酸度洛西汀对白念珠菌形态转换的影响;并通过二甲氧基苯基四氮唑盐(XTT)法和结晶紫染色法评估其对白念珠菌生物膜代谢活性及生物膜形成量的影响.结果 白念珠菌的最低抑菌浓度为15.625 µg/mL,最低杀菌浓度为31.25 µg/mL;31.25 µg/mL的盐酸度洛西汀可在第9小时有效杀死白念珠菌;31.25 µg/mL 和 15.625 µg/mL 的盐酸度洛西汀能阻止白念珠菌酵母向菌丝相转换,使白念珠菌保持酵母相.此外,盐酸度洛西汀对白念珠菌生物膜代谢活性和生物膜量同样具有抑制作用,且其抑制效果呈浓度依赖性.结论 盐酸度洛西汀在体外对白念珠菌具有显著抗菌效应.

Objective To systematically evaluate the effects of duloxetine hydrochloride on Candida albicans through in vitro experiments.Methods The minimum inhibitory concentration(MIC)and minimum fungicidal concentration(MFC)of duloxetine hydrochloride against the growth of C.albicans were detected by micro-broth dilution method;the time-fungicidal curve was used to examine the time-fungicidal trend of duloxetine hydrochloride against C.albicans.The effects of different concentrations of duloxetine hydrochloride on the morphological transformation of C.albicans were observed by inverted microscope;and the effects on the biofilm metabolic activity and biofilm formation of C.albicans were evaluated by the methyl tetrazolium salt(XTT)method and crystal violet staining.Results The MIC of C.albicans was 15.625 µg/mL,and the MFC was 31.25 µg/mL;31.25 µg/mL of duloxetine hydrochloride effectively killed C.albicans at 9 h;31.25 µg/mL and 15.625 µg/mL of duloxetine hydrochloride prevented the conversion of C.albicans from yeast to mycelial phase and allowed C.albicans to remain in yeast phase.In addition,duloxetine hydrochloride inhibited C.albicans biofilm metabolic activity and biofilm volume as well,and its inhibitory effect was concentration-dependent.Conclusion Duloxetine hydrochloride has significant antifungal activity against C.albicans in vitro.
