猪流行性腹泻病毒和猪德尔塔冠状病毒双重TaqMan荧光RT-PCR检测方法的建立与应用OA
Development and Application of a Duplex TaqMan Real-time RT-PCR for Porcine Epidemic Diarrhea Virus and Porcine Deltacoronavirus
本研究旨在建立一种能同时检测猪流行性腹泻病毒(Porcine epidemic diarrhea virus,PEDV)和猪德尔塔冠状病毒(Porcine deltacortonavirus,PDCoV)的双重TaqMan荧光RT-PCR方法.根据PEDV的N基因和PDCoV的M基因,设计合成特异性引物及TaqMan探针,通过反应条件和反应体系优化,建立快速检测PEDV和PDCoV的双重TaqMan荧光RT-PCR方法,对该方法的灵敏性和特异性进行评价.结果表明:该方法的检测灵敏度可达10 TCID50/mL;当PEDV和PDCoV病毒液效价在101~106 TCID50/mL范围内时,建立的标准曲线Ct值与病毒液浓度的常用对数呈现良好的线性相关;与猪传染性胃肠炎病毒(Transmissible gastroenteritis virus,TGEV)、猪轮状病毒(Porcine rotavirus,PRoV)、猪伪狂犬病毒(Pseudorabies virus,PRV)等常见病原体不发生交叉反应,具有很好的特异性.用建立的方法对2021-2022年收集的236份临床腹泻样品进行检测,PEDV阳性率45.76%(108/236),PDCoV阳性率14.41%(34/236),其中PDCoV和PEDV混合感染阳性率5.08%(12/236),本研究建立的双重TaqMan荧光RT-PCR方法可实现PEDV和PDCoV的同时鉴别检测,为实验室快速鉴别诊断猪病毒性腹泻提供了有效的技术支持.
The aim of this study was to develop a one-step duplex real-time RT-PCR assay to simultaneously detect and differentiate porcine epidemic diarrhea virus(PEDV)and porcine deltacoronavirus(PDCoV)so as to provide an efficient and sensitive tool for rapid diagnosis and epidemiological investigation of pig diarrhea.The specific primers and probes were designed for targeting to the conserved regions of PDCoV M gene and PEDV N genes.This duplex TaqMan real-time RT-PCR assay was developed and optimized for simultaneous detection of PEDV and PDCoV.The results showed this method had high specificity as it specifically detected PEDV and PDCoV without cross-reaction with TGEV,PRoV,PRV and other major porcine viruses.The lowest detection limit was 10 TCID50/mL for PEDV and PDCoV,indicating this assay was highly sensitive.In addition,236 clinical diarrhea samples collected between January 2021 and December 2022 were tested using this method and the positive rates were 45.76%for PEDV and 14.14%for PDCoV,in which the mixed infection rate was 5.08%.The results indicated that the established duplex qRT-PCR method provided a rapid,specific,sensitive and efficient technique for differential detection and epidemiological investigation of PEDV and PDCoV.
