猪圆环病毒2型和3型毒株的分离鉴定及致病性研究OA
Isolation,Identification,and Pathogenicity of PCV2 and PCV3 Strains
为了分离鉴定猪圆环病毒2型(Porcine circovirus 2,PCV2)、猪圆环病毒3型(Porcine circovirus 3,PCV3)以及确定其致病性.本研究利用实验室鉴定出的PCV2d、PCV3b阳性猪组织样本,分别用PK15细胞和实验室分离的猪肾上皮原代细胞分离获得PCV2和PCV3各1株,分别命名为PCV2JL11株和PCV3JL3株.经病毒滴度及病毒载量检测,PCV2JL11株病毒滴度为106 TCID50/mL,PCV3JL3株病毒载量4.638 lg(copies/μL).之后分别对小鼠和猪开展致病性试验.qPCR检测结果表明,PCV2和PCV3在小鼠和猪的各个脏器中均被检测到,并且伴有病毒血症.在动物回归试验中结果表明,攻毒PCV2株猪日增重为0.366 kg,与PBS组相比显著下降(P<0.05),攻毒PCV3组平均日增重为0.364 kg,与PBS组相比,有轻微下降,差异不显著(P>0.05).病理组织染色结果显示,PCV2/PCV3攻毒组肺肺泡壁增厚,炎症细胞数量减少,淋巴结淋巴细胞数量减少,排列疏松,界限模糊.本研究表明分离的PCV2JL11和PCV3JL3对断奶仔猪有致病性,为PCV2和PCV3的致病机制研究奠定基础.
To isolate and identify Porcine circovirus 2(PCV2)and Porcine circovirus 3(PCV3)and to assess their pathogenicity,we inoculated PCV2d and PCV3b-positive pig tissue samples from our laboratory onto PK15 cells and primary porcine kidney epithelial cells for viral isolation..Two virus strains were obtained and designated as PCV2JL11 and PCV3JL3.The PCV2JL11 strain exhibited a virus titer of 106 TCID50/mL and the viral load of the PCV3JL3 strain was measured at 4.638 lg(copies/μL).Subsequent pathogenicity tests were conducted in both mice and pigs.Quantitative PCR analysis confirmed the presence of PCV2 and PCV3 in various organs of both animal models.In animal experiments,pigs infected with the PCV2 strain exhibited an average daily weight gain of 0.366 kg,which was significantly lower than that of the PBS control group(P<0.05).In contrast,pigs infected with the PCV3 strain showed an average daily weight gain of 0.364 kg,which was not significantly different from the PBS group(P>0.05).Histopathological examination using hematoxylin and eosin staining revealed that the PCV2/PCV3 infection groups had thickness of lung alveolar walls,reduction of inflammatory cells,decrease of lymphocytes in the lymph nodes,and loose tissue arrangement with indistinct boundaries.Overall,these findings demonstrated that the isolated PCV2JL11 and PCV3JL3 strains were pathogenic to weaned piglets,thereby establishing a foundation for further investigations into the pathogenic mechanisms of PCV2 and PCV3.
