Prenylation of African Swine Fever Virus pA179L Protein and Its Effect on Apoptosis
蛋白质异戊二烯化是指将法尼基或香叶基异戊二烯基团共价连接到羧基末端含有CAAX基序的蛋白质上,其中C是一个不变的半胱氨酸,A1和A2通常是脂肪族残基,而X可以是任意氨基酸.本研究探讨了非洲猪瘟病毒(African swine fever virus,ASFV)编码的pA179L蛋白异戊二烯化修饰和抑制细胞凋亡之间的关系.结合PrePS程序预测和farnesyl抗体,证明了pA179L能够发生异戊二烯化修饰.本研究将pA179L的CNLI基序突变为SNLI或将其删除,然后比较其定位情况.应用核质分离、免疫印迹和间接免疫荧光分析发现异戊二烯化修饰调节pA179L定位.利用三种异戊二烯化抑制剂证明pA179L主要发生香叶基化修饰.本研究比较了野生型pA179L及其异戊二烯化钝性突变体pA179L-C176S对细胞凋亡的抑制作用和线粒体定位情况.结果显示,野生型pA179L能够通过异戊二烯化修饰靶向线粒体从而抑制细胞凋亡,而异戊二烯化钝性突变pA179L-C176S由于不能锚定线粒体而失去细胞凋亡抑制作用.本研究阐明了ASFV编码蛋白异戊二烯化修饰在调控细胞凋亡中的作用,为深入理解ASFV致病机制奠定了基础.
Protein prenylation refers to the covalent attachment of farnesyl or geranylgeranyl isoprenoid groups to proteins that have a CAAX motif at their carboxyl termini,where C is an invariant cysteine,A1 and A2 are usually aliphatic residues,and X can be any amino acid.This study investigated the relationship between prenylation modification of the African swine fever virus-encoded pA179L protein and its inhibition of apoptosis.Using the PrePS program prediction and farnesyl antibody,it was demonstrated that pA179Lhad undergone prenylation.In this study,the CNLI motif of pA179L was mutated to SNLI or deleted for comparison of their localization.Analyses of nuclear-cytoplasmic separation,immunoblotting and indirect immunofluorescence assays revealed that prenylation regulated pA179L localization.Three prenylation inhibitors were used to demonstrate that pA179L mainly undergwent geranylgeranylation.This study also compared the inhibitory effects on apoptosis and mitochondrial localization between wild-type pA179L and its prenylation-deficient mutant pA179L-C176S.The results showed that wild-type pA179L targeted mitochondria via prenylation to inhibit apoptosis whereas the prenylation-deficient mutant pA179L-C176S lost its apoptotic inhibition due to its inability to anchor to mitochondria.This study elucidated the role of ASFV-encoded protein isoprenylation modification in regulating apoptosis,laying a foundation for in-depth understanding of ASFV pathogenesis.
