阿尔茨海默病(AD)是一种渐进性发展神经退行性疾病,主要临床特征为记忆力减退、认知功能障碍、语言功能失调及后期行为能力丧失,因其发病机制复杂,尚无有效治疗药物.异常的表观遗传修饰与AD的发生发展密切相关,组蛋白甲基化是重要的表观遗传修饰之一,通过动态调控染色质结构和基因转录参与AD病理过程,对Aβ斑块形成、tau蛋白高磷酸化、神经炎症、突触可塑性及神经元死亡等发挥重要调节作用.中药在防治AD方面具有独特优势,研究表明中药及其有效成分可调控组蛋白甲基化改善AD,主要表现为减少Aβ与高磷酸化tau蛋白的生成、缓解神经炎症反应、修复突触可塑性损伤、增加神经元存活等.本文基于国内外研究现状对组蛋白甲基化在AD中的关键作用和中药及其有效成分干预研究进行综述,为中药的研发和应用提供新的思路.

Alzheimer's disease(AD)is a progressive neurodegenerative disorder characterized by memory loss,cognitive impairment,language dysfunction,and loss of behavioral abilities in its later stages.Because of its complex pathogenesis,there are currently no effective treatments available.Abnormal epigenetic modifications are closely associated with AD onset and progression of AD.Histone methylation is an important epigenetic modification that participates in AD pathology by dynamically regulating chromatin structure and gene transcription;it plays an important regulatory role in Aβ plaque formation,tau hyperphosphorylation,neuroinflammation,synaptic plasticity,and neuronal death.Traditional Chinese medicine has unique advantages for AD prevention and treatment;its active ingredients can regulate histone methylation to improve AD,mainly through reducing the production of A β and highly phosphorylated tau to alleviate neuroinflammatory responses,repair synaptic plasticity damage,and increase neuronal survival.This paper reviews the key role of histone methylation in AD and current research on traditional Chinese medicine interventions,providing a theoretical basis for the clinical treatment of AD and the research,development,and application of traditional Chinese medicine.
