目的 本研究探讨沉默JunB对C3H10T1/2细胞向棕色脂肪细胞分化及其产热能力的影响.方法 体外培养C3H10T1/2细胞,用小干扰RNA(siRNA)瞬时转染建立JunB沉默模型,经CCK-8和Western blot检测选取最佳JunB-siRNA组;采用鸡尾酒法诱导分化,分为空白对照(Control)组、诱导分化模型(DM)组、诱导分化模型+JunB沉默(DM+JunB-siRNA)组;通过油红O染色分析脂滴形成;Western blot和免疫荧光染色检测解偶联蛋白1(UCP1)和过氧化物酶体增殖物激活受体γ共激活因子1α(PGC-1α)蛋白表达;JC-1试剂盒检测线粒体膜电位.结果 JunB-siRNA对细胞活力无显著影响(P>0.05),JunB-siRNA Ⅰ沉默效果最佳(P<0.001);沉默JunB后棕色脂肪细胞脂滴积累增加,UCP1、PGC-1α表达上调(P<0.05,P<0.001),线粒体膜电位增强.结论 JunB沉默提高棕色脂肪细胞脂滴含量和产热能力,可能通过改善线粒体功能实现.

Objective We investigated the effect of silencing JunB on the differentiation of C3H10T1/2 cells into brown adipocytes and thermogenic capacity.Methods C3H10T1/2 cells were cultured in vitro and transiently transfected with small interfering RNA(siRNA)to establish a JunB silencing model.The best JunB-siRNA group was selected by Cell Counting Kit-8(CCK-8)and Western blot analysis.Differentiation was induced by a cocktail method,and blank control(Control),induced differentiation model(DM),and induced differentiation model with JunB silencing groups(DM+JunB-siRNA)were created.Lipid droplet formation was analyzed by Oil Red O staining.Western blot analysis and immunofluorescence staining were used to detect uncoupling protein 1(UCP1)and peroxisome proliferator-activated receptor γ coactivator-1α(PGC-1α)protein expression,and a JC-1 kit was used to detect mitochondrial membrane potential.Results JunB-siRNA had no significant effect on cell viability(P>0.05),with JunB-siRNA Ⅰ silencing having the best effect(P<0.001).JunB silencing increased lipid droplet accumulation in brown adipocytes.UCP1 and PGC-1α expression was up-regulated(P<0.05,P<0.001)and mitochondrial membrane potential was enhanced.Conclusions JunB silencing increased the lipid droplet content and thermogenesis of brown adipocytes,likely through improving mitochondrial function.
