目的 观察长蛇灸对"阳虚质"大鼠棕色脂肪组织(BAT)产热的影响,探索其改善阳虚质的机制.方法 15只SPF级SD大鼠随机分为正常组、模型组和长蛇灸组(每组5只).模型组和长蛇灸组通过趋热筛选及传代选育制备阳虚质模型,长蛇灸组造模后连续15 d接受长蛇灸治疗.检测各组肛温、体质量、趋热性、冰水游泳时间和自发活动次数;ELISA检测血清中促肾上腺皮质激素(ACTH)、皮质酮(CORT)和促肾上腺皮质激素释放激素(CRH)的水平;HE染色观察肩胛区BAT脂滴形态;免疫组化与Western blot法检测BAT中解偶联蛋白1(UCP1)、PTEN诱导激酶1(PINK1)、帕金森病相关蛋白(Parkin)表达.结果 与正常组相比,模型组肛温、体质量、冰水游泳时间和自发活动次数降低(P<0.01,P<0.001),趋热性增加(P<0.01),ACTH、CRH水平降低(P<0.01),BAT脂滴数量减少、空泡增大,且BAT中UCP1、PINK1、Parkin的蛋白表达均下调(P<0.001,P<0.000 1).与模型组相比,长蛇灸组治疗后肛温、体质量、冰水游泳时间和自发活动次数上升(P<0.05,P<0.01),趋热性降低(P<0.05),ACTH、CRH水平升高(P<0.01),BAT脂滴数量增多、空泡变小,UCP1、PINK1、Parkin水平升高(P<0.05,P<0.01,P<0.001,P<0.000 1).结论 长蛇灸可改善阳虚质大鼠症状和血清激素水平,其机制可能与激活PINK1/Parkin通路促进线粒体自噬,增加BAT中UCP1表达促进产热有关.

Objective To observe the effects of Long Snake Moxibustion on brown adipose tissue(BAT)thermogenesis in rats with a yang deficiency constitution,and explore the underlying mechanism involved.Methods Fifteen SPF-grade SD rats were randomly divided into normal,model,and Long Snake Moxibustion groups(five per group).The model and Long Snake Moxibustion groups underwent heat-preference screening and generational breeding to establish a yang deficiency constitution model.Following modeling,the Long Snake Moxibustion group received treatment for 15 consecutive days.Measurements included anal temperature,body weight,heat preference,ice-water swimming time,and spontaneous activity count.Serum levels of adrenocorticotropic hormone(ACTH),cortisol,and corticotropin-releasing hormone(CRH)were detected by enzyme-linked immunosorbent assay.Hematoxylin and eosin staining was used to observe lipid droplet morphology in interscapular BAT.The expression of uncoupling protein 1(UCP1),PTEN-induced putative kinase 1(PINK1),and Parkinson disease protein(Parkin)in BAT was detected by immunohistochemistry and Western blot analysis.Results Compared with the normal group,the model group exhibited decreased anal temperature,body weight,ice-water swimming time,and spontaneous activity(P<0.01,P<0.001),increased heat preference(P<0.01),lower ACTH and CRH(P<0.01),decreased BAT lipid droplet number with enlarged vacuoles,and downregulated protein expression of BAT,UCP1,PINK1,and Parkin(P<0.001,P<0.000 1).Compared with the model group,the Long Snake Moxibustion group showed increased anal temperature,body weight,ice-water swimming time,and spontaneous activity after treatment(P<0.05,P<0.01),decreased heat preference(P<0.05),elevated ACTH and CRH(P<0.01),increased BAT lipid droplet number with smaller vacuoles,and upregulated protein expression of BAT UCP1,PINK1,and Parkin(P<0.05,P<0.01,P<0.001,P<0.000 1).Conclusions Long Snake Moxibustion improved symptoms and serum hormone levels in rats with a yang deficiency constitution.The mechanism may be related to PINK1/Parkin signaling activation to promote mitophagy,increasing BAT UCP1 expression and enhancing thermogenesis.
