连翘苷调节JAK2/STAT3信号通路对大鼠痔术后创面愈合及炎症反应的影响及作用机制OA
Effects and mechanism of phillyrin on wound healing and inflammatory response after hemorrhoid surgery in rats via the JAK2/STAT3 signaling pathway
目的 探讨连翘苷(Phil)调节Janus激酶2(JAK2)/信号转导和转录激活因子3(STAT3)信号通路对大鼠痔术后创面愈合及炎症反应的影响.方法 采用冰醋酸注射法构建大鼠痔疮模型,并随机分为模型(Model)组、连翘苷低(Phil-L)、高剂量(Phil-H)组,连翘苷高剂量+香豆霉素A1(Phil-H+CA1)组,另取健康大鼠作为对照(NC)组,每组8只.游标卡尺测量并计算创面面积,观察并评估肛周水肿程度;全血细胞计数仪检测白细胞、中性粒细胞和淋巴细胞数;ELISA检测血清肿瘤坏死因子(TNF)-α、白细胞介素(IL)-6、IL-1β水平;苏木素-伊红(HE)染色观察肛周组织病理学变化;RT-qPCR检测肛周组织中JAK2、STAT3 mRNA水平;免疫印迹法检测肛周组织中血管内皮生长因子(VEGF)、基质金属酶(MMP)-9及JAK2/STAT3通路相关蛋白表达.结果 与Model组相比,Phil可以明显减少大鼠肛周组织中炎性细胞浸润,降低创面面积、肛周水肿程度,且白细胞数、中性粒细胞占比、JAK2、STAT3 mRNA水平、IL-1β、IL-6、TNF-α和p-JAK2、p-STAT3蛋白表达降低,淋巴细胞占比、VEGF、MMP-9蛋白表达显著升高(均P<0.05),且Phil-L、Phil-H组间差异显著(均P<0.05);与Phil-H组比,Phil-H+CA1组各定量指标均相反表达(均P<0.05).结论 Phil可以通过抑制JAK2/STAT3信号通路,抑制大鼠痔术后炎症反应,并促进创面愈合.
Objective To explore the effects of phillyrin(Phil)on wound healing and the inflammatory response after hemorrhoid surgery in rats by adjusting the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3)signaling pathway.Methods A rat model of hemorrhoids was established by acetic acid injection.Rats were divided randomly into Model,Phil-low-dose(Phil-L),Phil-high-dose(Phil-H),Phil-H+JAK2A activator coumermycin A1(CA1),and healthy control(HC)groups(n=8 rats per group).The wound area was measured using Vernier calipers,and the degree of perianal edema was observed and evaluated.The total blood cell count was used to detect the numbers of white blood cells,neutrophils,and lymphocytes.Serum levels of tumor necrosis factor(TNF)-α,interleukin(IL)-6,and IL-1β were detected by ELISA.Pathological changes in perianal tissues were observed by hematoxylin/eosin staining.JAK2 andSTAT3 mRNA in perianal tissues were detected by RT-qPCR and vascular endothelial growth factor(VEGF),matrix metalloproteinase(MMP)-9,and JAK2/STAT3 pathway-related proteins in perianal tissues were detected by immunoblotting.Results Compared with the Model group,Phil significantly reduced inflammatory cell infiltration in perianal tissue;decreased the wound area and perianal edema;reduced the white blood cell count,neutrophil percentage,JAK2,STAT3 mRNA and IL-1β,IL-6,TNF-α,phospho-JAK2,and p-STAT3 proteins;and increased the proportion of lymphocytes and VEGF and MMP-9 proteins(all P<0.05).The differences between the Phil-L and Phil-H groups were also significant(all P<0.05).All quantitative indicators showed opposite expression in the Phil-H+CA1 and Phil-H groups(all P<0.05).Conclusions Phil can inhibit the JAK2/STAT3 signaling pathway,suppress the inflammatory response after hemorrhoid surgery in rats,and promote wound healing.
