乌鲁木齐地区23例ABO亚型的三代测序分子遗传分析及新等位基因鉴定OA
Third-generation sequencing-based molecular genetic analysis and novel alleles identification of 23 ABO subtypes in Urumqi
目的 探讨乌鲁木齐地区 23 例含多民族的无偿献血者 ABO 亚型分子遗传背景.方法 筛选血清学试管法 ABO 正反定型不符的 ABO 亚型献血者标本共 23 例,采用 PCR-SSP 法或荧光 PCR(RT-PCR)法进行初步的ABO 基因分型,采用 PacBio 三代测序技术(third-generation sequencing,TGS)对所有标本进行 ABO 基因全长测序及单倍型分析.结果 23 例标本通过三代测序得到46 个具有ABO 等位基因单核苷酸变异(Single nucleotide variants,SNVs)的全长单倍型序列,获得常见已知 ABO 等位基因:ABO*A1.02、ABO*B.01、ABO*O.01.01、ABO*O.01.02,ABO*B102,已知 ABO 亚型等位基因:ABO*A2.01、ABO*AW.37、ABO*AW.25、ABO*AW.06、ABO*B3.01、ABO*BW.07 和 ABO*Bw34,并鉴定出 3 个 ABO 新等位基因:ABO*A2.NEW(c.1040_1041insA)、ABO*B.NEW(c.949A>G)、ABO*B.NEW(c.1029G>A),均已被 NCBI GenBank 收录并公布.结论 三代测序可以准确获得完整的 ABO 基因序列,区分单倍型,研究阐释了 23 例 ABO 亚型的分子遗传背景,为血型的准确鉴定和保障输血安全提供了参考依据.
Objective To investigate the molecular genetic background of ABO subgroups among 23 multi-ethnic volun-tary unpaid blood donors in Urumqi.Methods A total of 23 blood donor samples of ABO subgroups with ABO group dis-crepancies detected by serological tube test were collected.Preliminary ABO genotyping was performed by polymerase chain reaction with sequence-specific primers(PCR-SSP)or real-time fluorescence polymerase chain reaction(RT-PCR).PacBio third-generation sequencing(TGS)was then applied for full-length ABO gene sequencing and haplotype analysis for all samples.Results A total of 46 haplotype sequences containing single nucleotide variants(SNVs)of ABO alleles were obtained from 23 samples by TGS.Common known ABO alleles were identified including ABO*A1.02,ABO*B.01,ABO*O.01.01,ABO*O.01.02 and ABO*O.01.02.Known ABO subtype alleles included ABO*A2.01,ABO*AW.37,ABO*AW.25,ABO*AW.06,ABO*B3.01,ABO*BW.07 and ABO*BW34.Three novel ABO alleles were identified:ABO*A2.NEW(c.1040_1041insA),ABO*B.NEW(c.949A>G)and ABO*B.NEW(c.1029G>A),all of which have been deposi-ted and released in NCBI GenBank.Conclusion Third-generation sequencing can accurately obtain the complete sequence of ABO gene and distinguish haplotypes.This study elucidates the molecular genetic background of 23 ABO subtypes,provi-ding a reference for accurate blood group typing and ensuring transfusion safety.
