糕点中丙烯酰胺的超高效液相色谱串联质谱检测方法建立及膳食暴露风险评估OA
Establishment of UPLC-MS/MS Detection Method for Acrylamide in Pastries and Assessment of Dietary Exposure Risk
目的:建立固相萃取-超高效液相色谱串联质谱法测定糕点中丙烯酰胺(Acrylamide,AA)的分析方法,并评估其膳食暴露风险.方法:通过优化色谱条件、前处理等步骤,质谱多重反应监测模式(MRM)内标法定量建立适合检测糕点中AA的方法,采用点评估模型计算糕点中AA的暴露量.结果:在5~300 ng/mL线性范围内,AA具有良好的线性关系,相关系数(R2)0.999 89,检出限0.3 µg/kg,定量限1.1 µg/kg,回收率102.3%~105.3%,相对标准偏差(n=6)4.7%~8.4%;海南省市售糕点中AA残留量理论膳食暴露量低于ADI.结论:该方法前处理简单便捷,重现性好、准确性和灵敏度较高,适合大批量样品检测,满足实际检测要求.本次监测糕点中的AA残留量对人体健康的危害风险较低,不会对消费者健康产生危害,无需纳入风险管理程序.但为减少丙烯酰胺对健康的危害,仍需探索降低乃至可能消除AA的方法.
[Objective]To establish an analytical method for the determination of acrylamide in pastries using solid-phase extraction-ultra-high performance liquid chromatography-tandem mass spectrometry(SPE-UHPLC-MS/MS),and to evaluate its dietary exposure risk.[Method]A method suitable for detecting acrylamide(AA)in pastries was established through optimizing steps such as chromatographic conditions and sample pretreat-ment,with quantification performed via the internal standard method under the multiple reaction monitoring(MRM)mode of mass spectrometry.The point assessment model was used to calculate the dietary exposure of AA in pastries.[Result]The results showed that within the linear range of 5-300 ng/mL,AA exhibited a good linear relationship with a correlation coefficient(R2)of 0.999 89.The limit of detection(LOD)was 0.3 μg/kg,and the limit of quantification(LOQ)was 1.1 μg/kg.The recovery rate ranged from 102.3%to 105.3%,and the relative standard deviation(RSD,n=6)was 4.7%-8.4%.Additionally,the theoretical dietary exposure of AA in commercially available pastries in Hainan was lower than the acceptable daily intake(ADI).[Conclusion]This method features sample and convenient pretreatment,low interference,good reproducibility,high accuracy and sensitivity.It is suitable for large-batch sample testing and meets the requirements of practical detection.The monitoring results indicated that the health risk of AA in pastries from Hainan is relatively low,which will not cause harm to consumers'health and does not need to be included in risk management procedures.However,to reduce the health hazards of acrylamide,further research is still needed to explore methods for reducing or even potentially eliminating AA.
