一株豆粕发酵高产蛋白酶芽孢杆菌的筛选及特性研究OA

豆粕是大豆经压榨提取豆油时产生的一种副产物,年产量非常丰富.利用豆粕制成的大豆肽是一种更为优质的新型蛋白质改性产品,被广泛应用于制药、食品、化妆品、饲料制备及植物栽培等行业.目前利用微生物发酵法生产大豆肽,被认为是最为先进的方法之一.本研究从海参肠道内容物中筛选出一株高产蛋白酶芽孢杆菌菌株,并对其产蛋白酶条件及其豆粕液态发酵条件进行了优化.结果表明:该筛选菌株为一株贝莱斯芽孢杆菌(Bacillus velezensis),命名为芽孢杆菌DLH-1;在培养基成分为2%牛肉膏,1%酵母浸粉和1%NaCl,培养温度35℃,初始pH 7.0,接种量5%和培养时间33 h时,其产蛋白酶活力能够达到131.309 U/mL,比优化前提高了1.94倍;利用其在最优产酶条件下产生的发酵菌液对豆粕进行液体发酵时发现,在接菌量 4%,料水比 1:10,发酵水解时间 36 h,发酵水解温度 45℃,初始pH 6.0 时,对豆粕能够有效水解,产生的多肽含量比优化前提高 1.76 倍.本研究旨在为豆粕发酵制备蛋白饲料或肥料提供高性能菌源,同时为豆粕的开发利用及相关企业经济效益的提高等提供参考.

Soybean peptide made from soybean meal is a new protein modification product with better quality,which is widely used in pharmaceutical,food,cosmetics,feed preparation,and plant cultivation.At present,the production of soybean peptides by microbial fermentation is regarded as the most advanced method.In this study,a Bacillus strain with high protease production was selected from the intestinal contents of sea cucumber,and the conditions of protease production and liquid fermentation of soybean meal were optimized.The results showed that the strain was Bacillus velezensis(Bacillus DLH-1).When the medium composition was 2%beef paste,0.75%yeast powder and 1%NaCl,the culture temperature was 35℃,the initial pH 7.0,the inoculation amount was 5%and the culture time was 33 h,the proteinase-producing activity was 131.309 U/mL,which was 1.94 times higher than that before optimization.It was found that the soybean meal could be effectively hydrolyzed under the conditions of 4%inoculation amount,1:10 ratio of material to water,36 h fermentation hydrolysis time,45℃fermentation hydrolysis temperature and initial pH 6.0,and the polypeptide content was 1.76 times higher than that before optimization.The aim of this study was to provide high performance bacteria source for soybean meal fermentation to produce protein feed or fertilizer,and provide help for the development and utilization of soybean meal and the improvement of economic benefits of related enterprises.
