壳寡糖与羧甲基纤维素钠对热煮虹鳟鱼品质的影响及调控机制OA
Effects of Chitosan Oligosaccharides and Sodium Carboxymethyl Cellulose on Quality of Hot-boiled Rainbow Trout(Oncorhynchus mykiss)and Its Regulatory Mechanism
为探究热煮加工中虹鳟鱼肉质构劣变的调控策略,测定煮后虹鳟鱼肉的质构特性、保水性、微观结构、红外光谱以及肌原纤维蛋白含量、总巯基与羰基含量等蛋白质理化指标,系统比较壳寡糖(chitooligosaccharide,COS)和羧甲基纤维素钠(sodium carboxymethylcellulose,CMC-Na)对热煮虹鳟鱼品质特性及蛋白质稳定性的影响.结果表明,添加COS和CMC-Na均能改善鱼肉的质构特性(硬度、内聚性、弹性、咀嚼性等)及保水性,并通过抑制肌原纤维蛋白流失和增加蛋白质化学键稳定网络结构.COS主要通过其亲水性与自由水结合(红外光谱中PK1处的吸收峰强度减弱并发生位移),减少加热过程中水分流失,改善鱼肉的弹性和咀嚼性,同时促进蛋白质间的交联,抑制蛋白质氧化变性;CMC-Na可以在鱼肉表面形成致密的凝胶网络以锁住水分,同时通过增强蛋白质二硫键稳定蛋白质高级结构,减少蛋白的流失和变性,避免加热导致的鱼肉松散问题.研究结果揭示了以COS和CMC-Na为代表的多糖类添加剂通过差异化路径调控鱼肉热诱导变性的规律,为预制鱼制品加工中功能性配料的精准选用提供了理论依据.
In order to explore the regulation strategy of texture deterioration of rainbow trout meat during thermal cooking processing,the texture characteristics,water holding capacity,microstructure,infrared spectroscopy,myofibrillar protein content,total sulfhydryl and carbonyl content of cooked rainbow trout meat were determined.The effects of chitooligosaccharide(COS)and sodium carboxymethylcellulose(CMC-Na)on the quality characteristics and protein stability of hot-boiled rainbow trout were systematically compared.The result showed that the addition of COS and CMC-Na could improved the texture characteristics(hardness,cohesiveness,elasticity,chewiness,etc.)and water retention of fish meat,and stabilized the network structure by inhibiting the loss of myofibrillar protein and increasing protein chemical bonds.COS mainly combined with free water through its hydrophilicity(the absorption peak intensity at PK1 in the infrared spectrum weakened and shifted),reduced water loss during heating,improved the elasticity and chewiness of fish meat,promoted the cross-linking between proteins,and inhibited the oxidative denaturation of proteins.CMC-Na could formed a dense gel network on the surface of fish to lock water,and could stabilized the high-level structure of protein by enhancing protein disulfide bonds,reduced protein loss and denaturation,and avoided the looseness of fish caused by heating.Above results revealed that the polysaccharide additives represented by COS and CMC-Na regulated the heat-induced denaturation of fish meat through differential pathways,which provided a theoretical basis for the precise selection of functional ingredients in the processing of prefabricated fish products.
