响应面法优化宜昌蛇菰多糖提取工艺及柱前衍生HPLC法分析其单糖组成OA
Optimization of Extraction Process of Polysaccharides from Balanophora henryi Hemsl.by Response Surface Meth-odology and Analysis of Monosaccharide Composition via Pre-Column Derivatization HPLC
目的:优化宜昌蛇菰多糖的提取工艺,并分析其单糖组成,为蛇菰资源的开发利用提供实验依据.方法:在单因素试验基础上,以提取时间、温度和料液比为自变量,多糖得率为响应值,采用 Box-Behnken 试验设计结合响应面法优化水提醇沉工艺;利用 Sevage 法纯化多糖,经三氟乙酸水解后,采用 1-苯基-3-甲基-5-吡唑啉酮(PMP)柱前衍生高效液相色谱法对其含有的 9 种单糖组分进行定性与相对定量分析.结果:确定最佳提取工艺为料液比 1∶15、提取温度 90℃、提取时间 3 h,此条件下多糖提取率达 5.88%.单糖组成分析表明,宜昌蛇菰多糖由葡萄糖醛酸、甘露糖、核糖、乙酰氨基葡萄糖、氨基葡萄糖、葡萄糖、半乳糖、木糖、岩藻糖构成,其相对摩尔比为12.22∶4.54∶0.62∶2.01∶1.28∶36.61∶21.04∶10.29∶4.19.结论:所建立的响应面优化工艺稳定,PMP-HPLC单糖分析方法可行,适用于蛇菰多糖的提取及质量控制,为该天然产物的深入开发奠定了理论基础.
Objective To optimize the extraction process of polysaccharides from Balanophora henryi Hemsl.and analyze its monosaccharide composition,to provide experimental evidence for the development and utilization of B.Henryi resources.Methods Based on single-factor experiments,Box-Behnken design combined with response surface methodology was em-ployed to optimize the water extraction and alcohol precipitation process,with extraction time,extraction temperature and solid-liquid ratio as independent variables and polysaccharide yield as the response value.The crude polysaccharides were purified by the Sevage method,and after hydrolysis with trifluoroacetic acid(TFA),pre-column derivatization with 1-phe-nyl-3-methyl-5-pyrazolone(PMP)was performed,and high-performance liquid chromatography(HPLC)was used for qualitative and relative quantitative analysis of the nine monosaccharides.Results The optimal extraction conditions were determined as follows:solid-liquid ratio of 1∶15,extraction temperature of 90℃and extraction time of 3 h.Under these conditions,the polysaccharide yield reached 5.88%.Monosaccharide composition analysis indicated that the polysaccha-rides from B.henryi consisted of glucuronic acid,mannose,ribose,N-acetylglucosamine,glucosamine,glucose,galac-tose,xylose and fucose,with a relative molar ratio of 12.22∶4.54∶0.62∶2.01∶1.28∶36.61∶21.04∶10.29∶4.19.Conclusion The extraction process optimized by response surface methodology is stable,and the PMP-HPLC method for monosaccharide analysis is reliable.The established methods are suitable for the extraction and quality control of polysac-charides from B.henryi,which lay a theoretical foundation for further research and development of this natural product.
