黄连外泌体联合沙门氏菌外膜囊泡缓解结肠炎及其作用机制研究OA
Ameliorative Effects of Coptis chinensis Exosomes Combined with Salmonella Outer Membrane Vesicles on Colitis
目的:探讨黄连外泌体(CCEs)与沙门氏菌外膜囊泡(SOMVs)联合应用对 DSS 诱导小鼠结肠炎的改善作用及机制.方法:制备并表征 CCEs 及 SOMVs;建立 DSS 小鼠结肠炎模型,给予单药及联合干预.观察小鼠疾病活动指数(DAI)及病理变化,检测血清炎性因子、结肠组织肠道屏障蛋白及氧化应激指标.结果:提取的 CCEs 与SOMVs 均呈典型囊泡结构.与模型组及单药组相比,联合组对结肠炎症状及病理损伤的改善效果最佳(P<0.01),且血清 TNF-α、IL-6 和 IL-1β 等炎性因子水平显著降低,结肠 Claudin-1、Occludin 及 SOD 表达显著升高,MDA 降低.结论:CCEs 联合 SOMVs 可通过协同抗炎、修复肠道屏障及缓解氧化应激,有效改善小鼠结肠炎.
Objective To explore the ameliorative effect and mechanism of Coptis chinensis exosomes(CCEs)combined with Salmonella outer membrane vesicles(SOMVs)on dextran sulfate sodium(DSS)-induced colitis in mice.Methods CCEs and SOMVs were prepared and characterized.A mouse model of DSS-induced colitis was established,and the mice were treated with CCEs,SOMVs alone or their combination.The disease activity index(DAI)and pathological changes were e-valuated.Serum inflammatory factors,intestinal barrier proteins and oxidative stress indicators in colon tissues were detec-ted.Results Both CCEs and SOMVs exhibited typical vesicle structures.Compared with the model group and the monother-apy groups,the combined group achieved the optimal improvement on colitis symptoms and pathological lesions(P<0.01).The levels of serum TNF-α,IL-6 and IL-1β were significantly reduced.The expressions of Claudin-1,Occludin and SOD in colon tissues were markedly upregulated,while the content of MDA was downregulated.Conclusion The combina-tion of CCEs and SOMVs can ameliorate colitis in mice,and the mechanism involves synergistic anti-inflammation,intesti-nal barrier repair and attenuation of oxidative stress.
