仙鹤草化学成分鉴定及其抗血栓作用的网络药理学研究OA
Chemical constituent identification of Xianhecao(Agrimoniae Herba)and network pharmacology study on its antithrombotic effects
目的 分析仙鹤草乙酸乙酯部位和正丁醇部位的化学成分,并结合网络药理学与分子对接的方法,探究其抗血栓的作用机制.方法 采用超高效液相色谱-串联四极杆飞行时间质谱法(UPLC-Q-TOF-MSE)对仙鹤草乙酸乙酯部位和正丁醇部位进行定性分析;通过网络药理学筛选活性成分及靶点,获取血栓相关靶点,构建蛋白质-蛋白质相互作用(PPI)网络,进行基因本体论(GO)和京都基因与基因组百科全书(KEGG)富集分析,构建"成分-靶点-通路"网络,并通过分子对接验证关键成分与核心靶点的结合能力.结果 共鉴定出68个化合物,包括黄酮类、酚酸类、苯丙素类等.PPI分析显示白蛋白(ALB)、白细胞介素(IL)-6、肿瘤坏死因子(TNF)、蛋白激酶B1(AKT1)、表皮生长因子受体(EGFR)等为关键靶点.GO功能分析涉及细胞迁移、膜筏、蛋白激酶活性等;KEGG通路主要富集于血小板活化、补体与凝血级联、TNF、核因子(NF)-κB及磷脂酰肌醇3-激酶-蛋白激酶B(PI3K-AKT)等.筛选出14个核心成分(如儿茶素、槲皮素、山奈酚、咖啡酸等).分子对接结果显示,14个核心成分与关键靶点ALB、IL-6、TNF、AKT1、EGFR的结合能均低于-5 kcal/mol,结合活性良好.结论 仙鹤草中黄酮类、酚酸类和苯丙素类为主要活性成分,通过槲皮素、山柰酚、儿茶素等多成分作用于ALB、IL-6、TNF等多靶点,协同调控血小板活化、凝血级联及炎症等相关通路,从而发挥抗血栓作用.
Objective To analyze the chemical constituents of the ethyl acetate fraction and n-butanol fraction of Xianhecao(Agrimoniae Herba),and to explore its antithrombotic mechanism of action by combining network pharmacology and molecular docking approaches.Methods Ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MSE)was employed for qualitative analysis of the ethyl acetate fraction and n-butanol fraction of Xianhecao(Agrimoniae Herba).Network pharmacology was used to screen bioactive components and their corresponding targets,and thrombosis-related targets were obtained.A protein-protein interaction(PPI)network was constructed,followed by Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analyses.A"component-target-pathway"network was established,and molecular docking was performed to validate the binding affinity between key components and core targets.Results A total of 68 compounds were identified,including flavonoids,phenolic acids,phenylpropanoids,and others.PPI analysis revealed that albumin(ALB),interleukin-6(IL-6),tumor necrosis factor(TNF),protein kinase B1(AKT1),and epidermal growth factor receptor(EGFR)were key targets.GO functional analysis involved cell migration,membrane rafts,and protein kinase activity,among others.KEGG pathway enrichment was mainly concentrated in platelet activation,complement and coagulation cascades,TNF,nuclear factor-κB(NF-κB),and phosphatidylinositol 3-kinase-protein kinase B(PI3K-AKT)signaling pathways.Fourteen core components(e.g.,catechin,quercetin,kaempferol,and caffeic acid)were screened.Molecular docking results showed that the binding energies of the 14 core components with the key targets ALB,IL-6,TNF,AKT1,and EGFR were all below-5 kcal/mol,indicating favorable binding activity.Conclusion Flavonoids,phenolic acids,and phenylpropanoids are the main active components in Xianhecao(Agrimoniae Herba).Through multi-component actions of quercetin,kaempferol,catechin,and others on multi-targets including ALB,IL-6,and TNF,they synergistically regulate platelet activation,coagulation cascades,inflammation,and related pathways,thereby exerting antithrombotic effects.
