多方法结合探讨柑橘属中药材保健品的组方规律及对"气滞证"的作用机制OA
Multi-method investigation of formulation rules of citrus-based Chinese medicinal health products and their mechanisms of action on"qi stagnation pattern"
目的 通过数据挖掘、网络药理学和分子对接技术,探讨5种柑橘属中药材保健品的组方规律及对"气滞证"的作用机制.方法 收集化橘红、陈皮、枳壳、枳实及香橼相关保健品的组方信息,通过数据挖掘进行数据分析,通过网络药理学建立中药-疾病共同靶点PPI网络,对共同靶点进行GO及KEGG通路富集分析,并对核心靶点与中药有效成分进行分子对接进行验证.结果 组方中药材药性以温、平为主,药味以甘、苦为核心,"润肠通便""促进消化"为主要保健功能.对共同靶点进行GO及KEGG通路富集分析,共获取相关有效成分20个,共同靶点48个,得到核心靶点5个.GO功能富集分析得到条目449项,KEGG通路富集分析得到相关信号通路201条.5个核心靶点与它们对应的中药有效成分具有良好的亲和力.结论 柑橘属中药材所涉及的保健品组方多样,且其在对"气滞证"的作用方面,是一个多成分、多靶点、多通路参与的复杂过程.
Objective To explore the formulation rules of health products derived from five Citrus Chinese medicinal materials and their mechanisms of action on"qi stagnation pattern"through data mining,network pharmacology,and molecular docking techniques.Methods Formulation information on health products related to Huajuhong(Citri Grandis Exocarpium),Chenpi(Citri Reticulatae Pericarpium),Zhiqiao(Aurantii Fructus),Zhishi(Aurantii Fructus Immaturus),and Xiangyuan(Citri Fructus)was collected.Data mining was employed for analysis.A protein-protein interaction(PPI)network of common targets between the Chinese medicines and the disease was constructed using network pharmacology.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analyses were performed on the common targets.Molecular docking was then conducted to validate the binding between core targets and the active components of the Chinese medicines.Results The medicinal properties of the ingredients in the formulations were predominantly warm and neutral,with sweet and bitter flavors being the core tastes.The primary health functions were"moistening the intestines for bowel movement"and"promoting digestion."Enrichment analyses of the common targets identified 20 relevant active components,48 common targets,and 5 core targets.GO functional enrichment analysis yielded 449 items,while KEGG pathway enrichment analysis identified 201 related signaling pathways.The five core targets exhibited good binding affinity with their corresponding active components.Conclusion The health product formulations involving citrus Chinese medicinal materials are diverse.Moreover,their effects on"qi stagnation pattern"represent a complex process involving multiple components,multiple targets,and multiple pathways.
