剑叶玉簪总黄酮提取及活性研究OA
Extraction and Activity Study of Total Flavonoids from Hosta ensata F.Maekawa
以干燥粉碎的剑叶玉簪花为材料,采用单因素试验结合正交试验优化其总黄酮提取工艺,结果表明:剑叶玉簪总黄酮最佳提取工艺为料液比1∶40、60℃、20 min、90%乙醇,得率达(6.114±0.056)mg/g.进一步抗氧化实验显示,剑叶玉簪总黄酮对ABTS+·的清除效果显著优于 VC(p<0.05);在浓度较低时,对DPPH·和·OH 的清除能力优于 VC(p<0.05);铁离子还原能力在质量浓度≥0.1 mg·mL-1 时弱于VC(p<0.05),质量浓度≤0.05 mg·mL-1 时与 VC 无显著差异(p>0.05).运用CCK8 法检测剑叶玉簪花总黄酮对胃癌 BGC823 细胞的抑制作用,结果显示其对 BGC823 细胞具有明显抑制活性,IC50为23.84 μg·mL-1.综上,剑叶玉簪总黄酮展现出良好的抗氧化及抗肿瘤活性,具备潜在的应用价值.
Using dried and pulverized Hosta ensata flowers as the raw material,the extraction process for total flavonoids was optimized through single-factor experiments combined with orthogonal tests.Results demonstrated that the optimal extraction conditions were a solid-liquid ratio of1∶40,60℃,20 min,and 90%ethanol,achieving a yield of(6.114±0.056)mg/g.Further antioxidant assays revealed that the total fla-vonoids from Hosta ensata exhibited significantly stronger scavenging activity against ABTS+·compared to VC(p<0.05).At lower concen-trations,their DPPH·and·OH scavenging capacities also surpassed those of VC(p<0.05).However,their ferric ion-reducing power was weaker than VC when the concentration exceeded0.1 mg·mL-1(p<0.05)but showed no significant difference at concentrations≤0.05 mg·mL-1(p>0.05).The CCK-8 assay demonstrated that the total flavonoids significantly inhibited the proliferation of gastric cancer BGC823 cells,with an IC50 value of 23.84 μg·mL-1.In conclusion,the total flavonoids from Hosta ensata exhibit remarkable antioxidant and antitumor activities,highlighting their potential for further development in pharmacological or nutraceutical applications.
