Pullorum disease in chickens is an egg-transmissible and contact infectious disease caused by Salmonella pul-lorum.It can be transmitted vertically through hatching eggs,and horizontally via the digestive tract,mating,beak trimming,sex identification,and other routes.The main sources of infection are diseased chickens and carrier chickens.This disease not only leads to embryonic death during incubation,high morbidity and mortality in chicks,and decreased production performance of breeding chickens,but also poses a risk of zoonosis.The World Organization for Animal Health classifies it as a Class B infec-tious disease,and the Ministry of Agriculture and Rural Affairs of China has included it in key monitored diseases,explicitly re-quiring the positive rate of breeding chicken flocks to be controlled below 0.2%.Due to its simplicity,rapidity,high efficiency,low cost,and no requirement for serum separation,the whole blood plate agglutination test has become the preferred method for disease monitoring and purification quarantine in breeding chicken farms at the grassroots level.Aiming at the pain points in grassroots laboratory operations,this study systematically investigated the effects of key links such as whole blood sample pro-cessing,detailed test operations,and environmental condition control on test results.By optimizing operational parameters,clarifying graded judgment criteria,and identifying common error sources,a promotable standardized operational procedure was established.It provides a scientific and standardized operational basis for grassroots laboratories to conduct the whole blood plate ag-glutination test for pullorum disease,and effectively im-proves the quality of purification and quarantine of breeding chicken flocks.
